摘  要:目的:探讨在运动心脏的发生过程中,右心室肌蛋白质组的差异表达,筛选对研究运动心脏重塑机制有重要意义的右心室蛋白质。方法:18只雄性SD大鼠按运动能力和体重随机配伍分为对照组和运动组,每组9只。运动组进行12周中等强度有氧运动(70~80%VO2max),后与对照组同时称取体重、麻醉处死,称取心脏重量,提取右心室肌的全蛋白,对样品蛋白用双向凝胶电泳技术分离、后固定染色。运用Bio-rad PD quest图像分析软件对双向凝胶电泳图谱进行分析,选取差异蛋白进行质谱鉴定。结果:经过12周运动后,运动组心脏出现明显形态学变化。此次实验获得了重复性和分辨率较好的双向凝胶电泳图谱,与对照组相比,通过软件分析,上调到10倍以上或下调至1/10及以下的差异点31个,其中7个上调,24个下调。这些点最多位于分子量50~70kDa和10~20kDa、等电点7.0~9.0范围内。质谱分析并鉴定了其中2个蛋白点,包括丙酮酸脱氢酶E1α1(运动12周后下调)和一个未知蛋白。结论:12周运动后大鼠右室肌蛋白质组发生了明显变化。能量代谢酶的变化提示中等强度运动对心脏产生的重塑作用可能主要引起右心室肌能量代谢水平的变化。Objective To investigate the effect of exercise on the right ventricular proteome during the remodeling process of athletic heart. Methods Eighteen male SD rats were equally divided into two groups according to their athletic ability: the control group and the exercise group. After 12-week aerobic training (70-80%VO2max), they were narcotic and killed. Body weight and the heart were determined, and then the whole proteins of right ventricular muscle were extracted and then separated by two-dimensional gels electroghoresis (2 DE), and stained by coomassie brilliant blue dye. The protein concentration was measured by Bradford method. Several significant proteins were chosen to be identified. Results After 12-week training, significant morphological change and cardiac remodeling process appeared. Better reproducibility and resolution of two dimensional gel electrophoresis were obtained. As compared with the control group, 31 spots in which expression decreased less than 1/10 or increased over 10 folds were identified, including 7 up-regulated and 24 down-regulated. They were situated around Mr 50-70kDa/10-20kDa and pI 7.0-9.0, in which pyruvate dehydrogenase E1 alpha 1 (down-regulated after 12-week training) and an unknown 117kDa protein (up-regulated after 12-week training) were identified. Conclusion (1) During the remodeling process of athletic heart, proteome of the right ventricular muscle changed remarkably. (2) The change in metabolized enzyme indicated that the aerobic exercise could affect the energy metabolism level in myocardium.
