摘  要:目的研究人胃癌细胞系MGC-803经DNA甲基转移酶抑制剂5-氮-2-脱氧胞苷(5-Aza-CdR)和组蛋白去乙酰化酶抑制剂曲古抑菌素A(TSA)作用后细胞增殖和凋亡变化。方法MTT比色法测定各组细胞的增殖情况,Annexin V染色法检测各组细胞凋亡率。结果单独应用5-Aza-CdR或TSA作用于胃癌细胞系MGC-803后,细胞增殖均受到抑制,生长抑制率分别为68.0%、65.8%;凋亡率分别为(42.31±4.53)%、(39.30±5.37)%,而联合用药组细胞生长抑制率明显增加,达到86.3%,细胞凋亡率为(68.20±7.14)%。结论与单用5-Aza-CdR或TSA相比,联合应用两种药物更能显著的诱导细胞凋亡,抑制细胞增殖,这可能为胃癌的生物治疗提供一种新的思路。Objective To study the effects of the DNA methyltransferase inhibitor (5--Aza--2-- deoxycytidine, 5-- Aza-- cdR) and Histone deacetylase inhibitor(Trichostatin A, TSA) on cell proliferation and apoptosis in gastric cancer cell line MGC--803. Methods Cell proliferation was determined by MTT colorimetry and the rate of apoptosis was detected in each group by Annexin V staining. Results The growth of gastric cancer cell line MGC--803 was inhibited by 5--Aza--2--deoxycytidine or Trichostatin A and the inhibition rates were 68 %, 65.8 % and the apoptosis rates were 42.31 ±4.53 %, 39.30± 5.37 %. The inhibition rate and apoptosis rate in combination group were 86.3 % and 68.20 ± 7.14%. Conclusions Compared with 5--Aza--CdR or TSA,combination of the two drugs can more significantly induce apoptosis and inhibit cell proliferation, which may be a new idea to the biological treatment of gastric cancer.
