摘  要:[目的]通过诱变育种提高假丝酵母油脂的含量。[方法]以假丝酵母为出发菌种,利用紫外诱变、微波诱变及紫外和微波复合诱变分别对出发菌株进行诱变,筛选出生物量和吸光度均较大的菌株。[结果]酵母菌经紫外和微波诱变其生物量均比对照提高。在酵母产脂高产菌株的筛选中,微波与紫外线复合诱变是一种比较有效的方法,获得一高产酵母菌株Candida Yeast-1,其生物量比出发菌株提高率为43.3%,菌体油脂的吸光度比出发菌株提高率为32.1%。用索氏提取法测定其脂肪含量,脂肪含量提高了17.2%。[结论]对假丝酵母进行紫外、微波及复合诱变均能有效地提高其产脂率。[Objective] The aim was to increase the lipid content of candida through mutation breeding.[Method] With candida as original strain,the original strains were mutated by ultraviolet mutation,microwave mutation and compound mutation with ultraviolet and microwave mutation,resp.and the strains with bigger biomass and Absorbance were screened.[Result] Compared with the control,the biomass of strain treated by ultraviolet mutation and microwave mutation were all increased.During the screening of high yield strain of yeast producing lipid,the compound mutation with ultraviolet and microwave mutation was a more effective method and a high yield yeast strain of Candida Yeast-1 was obtained.The biomass of the strain was 43.3% higher than that of the original strain and the Absorbance of lipid of the strain was 32.1% higher than that of the original strain.Its fat content was detected by soxhlet extraction and the fat content was increased 17.2%.[Conclusion] The candida treated by ultraviolet mutation,microwave mutation and compound mutation with ultraviolet and microwave mutation all could increase its lipid producing rate.
