摘 要:目的本研究探索多次加温治疗对小鼠B16黑色素瘤疗效的相关治疗机制,比较加温次数对恶性黑色素瘤疗效的影响。方法实验采用自行开发的加热加湿生物实验平台,通过比较小鼠生存期,肿瘤重量,观察各组肿瘤的组织形态学变化,以及利用流式细胞术检测小鼠外周血中CD4+、CD8+、CD28+的表达来评价肿瘤治疗的效果。结果实验中各热疗组小鼠的生存时间要长于对照组(P<0.05),且以热疗3次组的小鼠生存期最长。各热疗组的肿瘤生长明显受到抑制(P<0.05),且热疗3次组的抑瘤效果最明显。组织病理学观察发现热疗组有大量肿瘤细胞呈凋亡、坏死样改变,热疗3次组瘤旁组织中有大量淋巴细胞浸润。此外同对照组比较各热疗组CD4+、CD8+、CD28+的表达均有升高(P<0.05)。结论多次热疗抑制肿瘤生长和激发机体免疫的效果要优于单次热疗。Objective In this study, in vivo investigations of the anticancer mechanism of hyperthermia treatment on mouse B16 malignant melanoma cells were carried out. Anti - cancer effect of multi - hyperthermia was also examined in vivo. Methods The in vivo test was performed on a home - made Biological Experimental Platform for hyperthermia by hot - humidified water vapor. The survival of mice and tumor weight were compared and histological examination was carried out. CD4^+ , CD8^+ and CD28^+ in peripheral blood were detected with flow cytometric analysis. Results Compared with the control group, groups under hyperthermia treatment had longer survival time, among which the group subjected to 3 times hyperthermia had longest survival time. Tumor growth was surpressed remarkably by hyperthermia and most noticeable effect happened to the 3 times treatment group. Histological examination revealed cell necrosis and apoptosis, including karyopyknosis, karyorrhexis, nuclear fragmentation and so on. Lymphocytic infiltration around tumor tissue was also observed in the mice received 3 times hyperthermia treatment. Conclusion Effect of multi - hyperthermia can greatly enhance the anti - tumor effect as compared with single treatment due to the considerations of tumor suppression and the anti - cancer immunity induction.
