虎杖提取液干预对NAFLD大鼠脂肪组织相关基因表达的影响

摘　　要：目的在基因表达水平上分析虎杖提取物对非酒精性脂肪肝(NAFLD)大鼠模型的干预效果及其作用机制。方法以高脂饲料饲养Wistar大鼠诱发NAFLD,然后转以基础饲料饲养,并对NAFLD大鼠进行虎杖提取液干预与对照实验。通过RT-qPCR方法检测干预组和对照组大鼠脂肪组织瘦素、脂联素、肿瘤坏死因子α、抵抗素和解偶联蛋白2的mRNA相对水平,以t检验分析两组间数据的差异。结果干预组大鼠脂肪组织瘦素mRNA和脂联素mRNA相对水平比对照组有所下降(P>0.05;P>0.05)。干预组大鼠脂肪组织的肿瘤坏死因子αmRNA相对水平比对照组显著下降(P<0.05)。所有对照组样品均可检测到解偶联蛋白2mRNA,而只有个别干预组样品可检测到较低水平的解偶联蛋白2mRNA。干预组和对照组样品均未检测到抵抗素mRNA。结论虎杖是TNF-α的抑制药物,可有效改善肝组织的脂肪变性和炎症的状况。Objective To detect the intervenient effect of the extracts of Polygonum caspidatum for the NAFLD rats and analyze its mechanism at the level of gene expressions. Methods The male Wistar rats were fed with highfatty chow to induce NAFLD. And then,these two groups of rats were fed with the low fatty chow and separately intervened with the extracts of Polygonum caspidatum or controlled with distill water. The mRNA relative level of leptin, adiponectin, tumor necrosis factor α, resistin and uncoupling protein 2 was detected in RT - qPCR. The data differences between two groups were analyzed in t - test, Results Both leptin and adiponectin mRNA in the intervenient group were separately lower than that of control group（ P 〉0. 05 ;P 〉 0.05 ）. TNF -α mRNA in the intervenient group was significantly decreased to that of the control group （P 〈 0.05 ）. UCP2 mRNA of all samples in the control group could be detected and few samples in the intervenient group were in the lower level of UCP2 mRNA. In all samples, their resistin mRNA didn＇t been detected. Conclusion Polygonum caspidatum is the TNF - α inhibitor. It can improve the steatosis and inflammatory reaction in liver tissue.
