摘 要:目的:研究奈达铂(NDP)联合顺铂(DDP)对人卵巢癌细胞株Skov-3增殖及凋亡的影响,并探讨其抑制增殖及诱导凋亡的机制。方法:采用MTT法检测不同浓度NDP与DDP单独或联合应用对Skov-3细胞的增殖抑制率;采用流式细胞术分析早期凋亡细胞比例;采用RT-PCR和Western blotting法分析增殖相关基因(蛋白)Ki-67及凋亡相关基因(蛋白)Bax、Bcl-2表达的变化。结果:NDP、DDP单独应用及两者联合使用明显抑制Skov-3细胞的生长,且呈剂量依赖性;根据中效原理发现两者联合使用高浓度时呈协同效应;流式细胞术结果表明正常对照组、NDP组、DDP组和联合组细胞早期凋亡比例依次增加;与单独用药组比较,联合用药组Ki-67、Bcl-2基因(蛋白)表达率显著降低,而Bax基因(蛋白)表达显著增高。结论:NDP和DDP联合应用与两药单独应用相比,对Skov-3细胞增殖的抑制作用和诱导凋亡能力均有明显增强。Objective:To study the synergistic effects of nedaplatin(NDP) and cispahin(DDP) on the human ovarian carcinoma Skov-3 cell line in vitro. Methods : The inhibition effects were evaluated by MTT assay. Cell apoptosis was detected by flow cytometry. The changes of Ki-67, Bax and Bcl-2 in mRNA and protein level were quantified by RT-PCR and Western blotting. Results : The growth inhibition of Skov-3 was dose-dependent after exposure to the NDP or DDP alone. The interaction of the two drugs was synergistic at higher concentrations according to the median-effect principle. The cells apoptotic rate was tended to increase. Compared with NDP or DDP treatment group, there was a decrease in Ki-67 and Bcl-2 mRNA(protein) expression in the combinative group; but an increase in the expression of Bax mRNA (protein) was observed. Conclusion: Compared with the effects of using high concentration NDP and DDP alone, the method of combining low concentration NDP and DDP proves to be much more effective in the inhibition of the proliferation and the induction of the apoptosis of Skov-3 cell line in vitro.
