摘 要:从运行稳定的能同步脱臭的曝气生物滤池中采集样品,富集分离获得一株能高效脱硫脱氨氮的菌株TS-1。对分离菌株进行形态观察、生理生化试验及16S rRNA基因序列分析,结果表明,该菌株TS-1为革兰氏阳性菌,杆状;菌落在营养琼脂培养基上呈圆形,表面光滑,乳白色半透明;V-P试验阴性,能水解淀粉和明胶,利用柠檬酸盐生长;对菌株进行16S rRNA的PCR扩增,扩增产物测序结果表明分离菌株TS-1与巨大芽孢杆菌(Bacillus megaterium)同源性达到99%;以16S rRNA同源性为基础构建了包括24株相关种属细菌在内的系统发育树,在系统发育树中,分离菌株TS-1与Bacillusmegaterium在同一分支。结合形态观察、生理生化试验及16S rRNA基因序列分析结果,将其初步鉴定为巨大芽孢杆菌(Ba-cillus megaterium)。在常温(30±2)℃、转速为150 r/min的条件下,处理pH7、S2-和NH4+-N分别为80 mg/L和88 mg/L的水样40 h,硫化物和氨氮的脱除率分别为91.8%和96.6%。A bacteria TS-1 with desulfurization and ammonia-nitrogen reduction was obtained by separation and purifying from sludge in a BAF with the capacity of deodorizing. The results of morphology, physiological and biochemical test together with 16S rRNA genetical sequence analysis indicated that: the cells of the strain TS-1 were Gram positive, straight rods; the colonies were roughly round, smooth, translucent and milk white in color in nutrient agar medium; V-P negative, it could utilize citrate , hydrolyze the starch and liquefy the glutin; analysis of partial 16S rRNA gene sequencing demonstrated that TS-1 belonged to the branch of Bacillus megateriurn, with a similarity of 99% ; a phylogenetic tree was constructed by comparing with the published 16S rRNA sequence of relative bacteria species, TS-1 and Bacillus megaterium constitute a branch in the phylogenetic tree. The results indicated that strain TS-1 was belonged to Bacillus megaterium. Under the condition of (30 ± 2) ℃ and 150 r/min, TS-1 had desulfurization capability removal rate of ammonia-nitrogen was 96.6% when of 91.8% when dealing with 80 mg/L sulfion 40 h, and the dealing with 88 mg/L ammonium-nitrogen.
