摘 要:[目的]探讨由伽玛刀照射所诱发的急性放射性脑水肿大鼠血管内皮生长因子(VEGF)表达的变化及其与放射性脑水肿的关系。【方法】单次50Gy伽玛刀照射大鼠大脑半球中线右旁开2mm(中心点在双外耳道连线上)建立急性放射性脑水肿模型,用干湿重法和免疫组织化学方法检测照射后不同时间段脑含水量和VEGF的表达。【结果】伽玛刀照射后12h脑含水量和靶区、靶周围区VEGF表达增加,照射后2d达到高峰,14d后仍有表达,VEGF表达的阳性细胞数和脑含水量呈显著正相关(r=0.9732,P〈0.05)。【结论】VEGF和急性放射性脑水肿的形成密切相关。[Objective] To study the relation between the expression of vascular endothelial growth factor (VEGF) and acute radiation-induced cerebral edema after Gamma knife irradiation. [Methods] The experi- mental model was established in rats by irradiating 2mm right to median line in cerebral hemisphere with 50Gy Gamma knife(the point of centricity lies in the line of auditory canal). The changes of brain water content (BWC) were measured by the wet and dry weight methods. Immunohistochemistry was used to determine the expression of VEGF during different periods after Gamma knife irradiation. [Results]BWC and the expression of VEGF in target area and peripheral zone were increased after 12h by Gamma knife irradiation and achieved peak after 2d. The expression of VEGF in target area and peripheral zone was still higher than normal after 14d. The positive ceils of expression of VEGF were positively with BWC (r=-0. 9732, P〈0.05). [Conclusion]VEGF is closely related with acute radiation- induced cerebral edema.
