摘  要:目的观察病程3个月、6个月链脲佐菌素(streptozocin,STZ)诱导的糖尿病大鼠视网膜毛细血管超微结构的病理改变及血管生成素-1(angiogenin,Ang-1)对超微结构改变的影响。方法应用STZ制备SD大鼠糖尿病模型。将SD大鼠30只随机分为糖尿病3个月组(DM3组)、糖尿病6个月组(DM6组),Ang-1治疗3个月组(ANG3组)、Ang-1治疗个月组(DM3组)、糖尿病6个月组(DM6组),Ang-1治疗3个月组(ANG3组)、Ang-1治疗6个月组(ANG6组)及正常对照3个月组(CON3组)、正常对照6个月组(CON6组)。尾静脉一次性注射STZ(55mg·kg-1)诱发大鼠糖尿病。ANG组大鼠每只眼球每15d球后注射Ang-1溶液0.05mL(0.2μg Ang-1)。每组大鼠各5只,分别饲养到3个月、6个月时处死,取大鼠视网膜进行电镜观察。结果(1)视网膜毛细血管超微结构:CON组内皮细胞和周细胞内异染色质分布均匀,细胞器、细胞核形态正常,基底膜结构清楚,连续完整。DM组内皮细胞水肿,线粒体肿胀、变性,核染色质浓缩、边集,分布于核膜下;基底膜明显增厚。ANG3组血管壁、内皮细胞核及周细胞核形态正常;ANG6组血管壁完整无断裂,基底膜增厚不明显。(2)组间视网膜毛细血管基底膜厚度均数比较:ANG3(228.0±16.2)nm与DM3(245.5±11.9)nm比较,差异有统计学意义(P<0.05);ANG6(251.5±19.3)nm与DM6组(310.6±28.0)nm比较,差异有显著统计学意义(P<0.01)。结论Ang-1对STZ糖尿病大鼠视网膜毛细血管超微结构的损害有明显防治作用。Objective To observe the pathologic changes of ultrastructure of retinal capillary in diabetic rats caused by streptozocin(STZ) at the course of disease for 3 months and 6 months,and to evaluate the effect of angiogenin-1 (Ang-1) on ultrastructural changes. Methods Animal models were induced by STZ(55 mg · kg^-1 mass) with one time injection at vena candalis on 30 Sprague-Dawley rats, and which were randomly divided into diabetic rats for 3 months group ( DM3 group ), 6 months group (DM6 group), diabetic rats treated with Ang-1 for 3 months group (ANG3 group), 6 months group ( ANG6 group), control group for 3 months ( CON3 group ) , 6 months ( CON6 group) ,5 rats in each group. Rats of ANG groups were injected intraocularly with Ang-1 0.05 mL(0.2 μg Ang-1 ) every fifteen days. The rats were killed after 3 months and 6 months, respectively, and their retinas were studied by electronic microscope. Results ( 1 ) About the retinal blood capillary ultrastructure, in the CON groups, the heterochromatine distributed evenly in both retinal blood capillary endothelial and perithelial cells, the shapes of organelles and nucleus were normal, the structure of retina capillary basement membrane displayed clear and integrity;In the DM groups, there were edema of endothelium, swelling and degenerated mitochondrion, limited nuclear chromatin in retinal blood capillary, and basement membrane was obviously thickened; In the ANG3 group, the structure of vessel wall was integrity, the nucleus of endothelial and perithelial ceils were normal;In the ANG6 group, the vessel wall was integrity without break, basement membrane was normal. ( 2 ) About the retinal capillary basement membrane thickness results of each group, there was statistical significance between ANG3 group (228.0 ± 16.2 ) nm and DM3 group (245.5 ± 11.9 ) nm (P 〈 0.05 ), and also between DM6 group ( 310.6 ± 28.0 ) nm and ANG6 group ( 251.5 ± 19.3 ) nm ( P 〈 0.01 ).
