摘  要:比较了三氯乙酸/丙酮沉淀法、酚抽法、丙酮沉淀法3种方法对香蕉(M.AAA Group Cavendish)果实总蛋白的提取效果。结果显示酚抽法所得图谱条带完整清晰;TCA法所得图谱条带间模糊不清,存在拖尾现象;而丙酮沉淀法提取的蛋白得率不高,蛋白带谱不完整。说明酚抽法适合香蕉果实总蛋白提取。同时为满足不同激素处理及采后不同成熟度香蕉果实高质量蛋白质提取的需要,在酚抽法基础上对提取缓冲液进行了改良,获得了良好的效果。为以后研究香蕉果实蛋白质功能奠定了基础。Three methods of total protein extraction from banana (Musa AAA Group Cavendish)fruit,including trichloroacetic acid/acetone precipitation method, phenol extraction method and acetone precipitation method were compared in this paper. The results showed that the spectrum of proteins extracted with phenol extraction method was complete and clear; while that obtained with trichloroacetic/acetone precipitation was blurred and smearing,and using acetone precipitation resulted in low protein recovery and the protein bands were fewer, so it showed that the phenol extraction was suitable for extracting total protein from banana fruit. With improved extraction buffer solution ,this method achieved a good result when used to extract proteins from banana fruit treated with different hormones. This study provides a useful method for banana fruit protein research.
