实时荧光定量RT-PCR检测结肠癌中Musashi-1 mRNA的表达

摘　　要：目的:了解正常结肠干/祖细胞标志Musashi-1mRNA在结肠癌中的表达情况。方法:应用实时荧光相对定量RT-PCR的方法检测25例结肠癌组织和18例远端正常结肠组织中Musashi-1mRNA的表达,用2-△△CT的方法计算相对于远端正常组织结肠癌组织中Musashi-1mRNA的表达倍数。结果:经实时荧光定量RT-PCR法检测,结肠癌组织中Musashi-1mRNA的表达与远端正常结肠组织相比有差异(P<0.01),且是远端正常结肠组织的4.24倍(1.95～9.23)。低分化组织与高、中分化组织相比,Musashi-1mRNA的表达有差异(分别为P<0.001、P<0.005)。低分化组织中Musashi-1的表达是高分化组织的5.54倍(3.07～9.98)。结论:正常肠干/祖细胞标志物Musashi-1在结肠癌组织中尤其是低分化组织中的高表达提示Musashi-1可能在结肠肿瘤的发生发展中起着一定的作用。Objective：To explore the expression of Musashi-1 mRNA in colonic cancer,which is a putative intestinal stem and early progenitor cell marker. Methods：The mRNA expression of Musashi-1 gene in 25 colonic cancer specimens and 18 distal normal colon tissues was detected by relatively quantitative real-time RT-PCR method. The multiples of expression of Musashi-1 in colonic cancer relative to distal normal colon tissue was calculated by the method of 2^-△△CT. Results：By Real-time RT-PCR method,the expression of Musashi-1 mRNA in colonic cancer was 4.24 （1.95-9.23）times higher than that in distal normal colon tissue. There was a significant difference between them（P 〈 0.01 ）, The expression of Musashi-1 mRNA in the poorly differentiated colonic cancer was signif- icantly different from the high and medium differentiated tissues （P 〈 0.001 and P 〈 0.005）. The expression of Musashi-1 mRNA in the poorly differentiated colonic cancer was 5.54（ 3.07-9.98 ）times higher than that in the high differentiated tissues. Conclusion：The expression of Musashi-1 mRNA in colonic cancer especially is higher in poorly differentiated tissues than in distal normal colon tissue, which suggests that Musashi-1 ,the intestinal stem and early progenitor cell marker,may play an important role in the initiation and development of the colonic cancer.
