摘  要:目的:模拟高原条件下大鼠牙周炎模型牙周组织中肿瘤坏死因子-α(TNF-α)、前列腺素E2(PGE2)的表达情况。方法:将80只SD大鼠随机分为缺氧对照组、缺氧实验组、常氧对照组及常氧实验组,每组20只,成功建立大鼠牙周炎模型(缺氧实验组、常氧实验组)后,在常氧、低氧环境(模拟海拔5000m高原、23h/d)下饲养,8周后处死动物,取牙周组织标本作切片,观察牙周组织病理变化状况,并采用免疫组织化学SABC法检测TNF-α、PGE2水平变化。结果:缺氧对照组与常氧实验组牙周组织中TNF-α、PGE2表达无显著差异,其余各组间牙周组织中TNF-α、PGE2表达情况不同(P<0.05)。结论:低压、低氧条件下SD大鼠牙周组织中TNF-α、PGE2较常氧条件下明显升高。AIM: To investigate the expression of tumor necrosis factor-α( TNF -or)and prostaglandin E2 (PGE2 ) in periodontal tissues of a rat model with periodontitis exposed to hypoxia at high altitude. METHODS: Eighty adult SD rats were divided randomly into 4 groups: the hypoxia group( n = 20), the hypoxia control group( n = 20), the normoxia group(n =20) and the normoxia control group(n =20). Ttwo successful rat periodontitis model groups(the hypoxia group and the normoxia group)were raised under hypoxia (simulating the altitude 5000 meters, 23 h per day) and normoxia environment separately for 8 weeks and then the rats were sacrificed and analyzed. Pathological assay was used to detect the general conditions and pathological changes of rat periodontal tissues. Immunohistochemical staining was conducted to determine the expressions of TNF -α and PGE2 in periodontal tissues in different groups of rat model. RESULTS : The levels of TNF -α and PGE2 in the the hypoxia group were not significantly higher than those in the normoxia control group( P 〉 0.05 ). The expressions of TNF -α and PGE2 in the other groups were significantly different ( P 〈 0.05 ). CONCLUSION : The levels of TNF -α and PGE2 were increased under hypoxia environment at high altitude.
