金属蛋白酶-2/金属蛋白酶组织抑制因子2比值与肝细胞癌侵袭力的关系研究

摘　　要：目的:探讨金属蛋白酶-2/金属蛋白酶组织抑制因子2、非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2比值与肝细胞癌侵袭力的关系。方法:利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳酶谱和反向酶谱分析技术对32例肝细胞癌组织中金属蛋白酶-2和金属蛋白酶组织抑制因子2含量及非活化的金属蛋白酶-2前酶/金属蛋白酶组织抑制因子-2,金属蛋白酶-2/金属蛋白酶组织抑制因子2比值进行检测并对比分析。结果:金属蛋白酶组织抑制因子2的分子量为18 000;癌旁组织中非活化金属蛋白酶-2的前酶/金属蛋白酶组织抑制因子2(0.94),金属蛋白酶-2/金属蛋白酶组织抑制因子2(0.42)均<1.0;肝细胞肝癌组织中非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2(1.70),金属蛋白酶-2/金属蛋白酶组织抑制因子2(1.64)均>1.0;转移组的2项比值也明显高于无转移组(P<0.01)。结论:非活化金属蛋白酶-2前酶/金属蛋白酶组织抑制因子2,金属蛋白酶-2/金属蛋白酶组织抑制因子2的异常变化可能是评价肝细胞癌生物学行为的有力指标。Objective To investigate the relationship between the level of MMP-2 and TIMP-2 and the invasion of hepatocelluar carcinoma. Methods MMP-2 and TIMP-2 were isolated from tissues of 32 cases of hepatocelluar carcinoma. The activities of MMP-2 and TIMP-2 were determined with Zymogaphy and reverse Zymogaphy, respectively. Results There were statistic differences of the activities of MMP-2 and TIMP-2 between the tissues of hepatocellular carcinoma and normal tissues （P〈0.05）. The ratio of PreMMP-2/TIMP-2（1. 70）, MMP-2/TIMP-2（1. 64） in malignant tissues was higher than those（0. 94,0. 42） in normal tissues, meanwhile, the ratio of PreMMP-2/TIMP-2 （1.97）, MMP-2/TIMP-2 （2. 20） in malignant tissues with metastasis was higher than those （1. 70, 1.64） in malignant tissues without metastasis. Conclution There are correlations between PreMMP-2/ TIMP-2, MMP-2/TIMP-2 and malignancy of hepatocellular carcinoma. The ratios of PreMMP-2/ TIMP-2 and MMP-2/TIMP-2 mighe be used as the potential indexes for the prediction of the biological behaviors of hepatocellular carcinoma.
