摘  要:目的观察E-64-D对钙蛋白酶(Calpain)表达的影响,探讨其促进脊髓恢复的作用机制。方法制作大鼠脊髓损伤动物模型(n=40),分为2组:脊髓损伤组(组1,n=20),E-64-D治疗组(组2,n=20)。应用Western blot技术检测Calpain表达变化情况。应用免疫组化技术观察Calpain、MAP2以及GFAP表达,并进行神经组织凋亡(TUNEL法)检测。采用BBB评分观察神经恢复情况。结果E-64-D治疗组Calpain表达下降,GFAP阳性细胞数减少,MAP2阳性细胞数增高,凋亡细胞减少,BBB评分评分增加。结论E-64-D通过抑制Calpain减少细胞凋亡抑制MAP2降解和胶质增生从而促进脊髓损伤的恢复。Objective To evaluate the neuroprotective effects of E-64-D in a rat spinal cord injury model.Methods 40 Wistar rats SCI models were made.The rats were randomly divided into two groups: trauma control group(group 1, n = 20) and E-64-D treatment group (group 2, n = 20). The activity of Calpain were examined via Western blotting. The expression of MAP2 and GRAP were detected by immunohistochemical technique respectively. The level of cells apoptosis was detected by TUNEL methods. Neurological performance was assessed using BBB(Basso, Beatlie, Bresnahan score)system. The results were analysed statistically. Results E-64- D can inhibit the activity of Calpain, reduce apoptosis, decrease GFAP expression, increase MAP2 expression, improve BBB scores. Conclusion E-64-D can reduce Calpain activity, improve functional outcome by means of reducing apoptosis and decreasing GFAP expression and inhibiting MAP2 degradation.
