C2-神经酰胺对牛晶状体上皮细胞凋亡的影响

摘  要:目的:观察C2-神经酰胺对牛晶状体上皮细胞(lens epithelial cells,LECs)凋亡的影响及半胱天冬酶-3(Caspase-3)活性的变化。方法:取第二代牛LECs,经不同浓度(25μmol/L,50μmol/L,100μmol/L)C2-神经酰胺作用12h,行TUNEL染色,荧光显微镜下计数凋亡细胞,计算凋亡率;Caspase-3活性检测试剂盒检测牛LECs中Caspase-3活性的变化。结果:随着C2-神经酰胺浓度的增加,牛LECs凋亡率也随之升高(P<0.05),呈剂量依赖关系。随着C2-神经酰胺浓度的增加,牛LEC中Caspase-3活性的表达也随之增强(P<0.05)。结论:C2-神经酰胺可以诱导牛LECs发生凋亡;在此过程中,存在Caspase-3活性表达。Aim: To study the effects of C2-ceramide on the apoptosis of cattle lens epithelial cells(LECs) , and observe the variation of Caspase-3 activity. Methods:The second generation LECs were treated with different concentrations (25 I.Lmol/L, 50 μmol/L, 100 μmol/L) of C2-ceramide for 12 h. After TUNEL staining,the rate of cell apoptosis was calculated; the variation of Caspase-3 activity was determined by Caspase-3 activity assay kit. Results :Cell apoptosis rate and the expression of Caspase-3 activity both increased with the increase of concentrations of C2-eeramide in a dose-dependent manner( P 〈 0. 05 ). Conclusion : C2-ceramide could induce apoptosis of LECs, and there is the expression of Caspase-3 activity in this process.
