黄芩苷对博莱霉素诱导的肺纤维化的影响

摘　　要：目的:观察黄芩苷(Bai)对博莱霉素(BLM)诱导的肺纤维化的影响,并探讨了可能的机制。方法:68只雄性SD大鼠,随机分为以下4组:BLM+Bai组、BLM+生理盐水(NS)组、NS+Bai组和NS+NS组。气管内一次性滴注BLM(5 mg.kg-1bw)或等体积NS的当天腹腔注射Bai(12.5 mg.kg-1.d-1,×28 d)或等体积NS。检测肺羟脯氨酸含量、肺胶原纤维阳性面积、肺Ⅰ型前胶原[ColⅠ(α)]mRNA表达、肺间质肌成纤维细胞数量。结果:与对照大鼠相比,气管内滴注BLM后第28 d,大鼠的肺组织羟脯氨酸含量增高;肺间质胶原纤维阳性面积增大;肺内ColⅠ(α)mRNA含量增高;肺间质肌成纤维细胞数量增多(均P<0.01)。上述指标的异常变化均在腹腔注射Bai后有所改善(均P<0.05)。结论:Bai有阻止BLM诱导肺纤维化的作用;Bai的上述作用与其阻止肺内Ⅰ型胶原异常合成和阻止肺间质肌成纤维细胞数量增多有关。Aim： To explore the effect of baicalein （Bai-Chinese Traditional Medicine） on bleomycin（BLM）-induced fibrosis in lungs of rats and its possible mechanism. Methods： Sixty-eight male Sprague-Dawley rats were randomly divided into 4 groups： BLM plus Bai group, BLM plus normal saline（NS） group, NS plus Bai group, and NS plus NS group. The rats were received single intratracheal instillation of BLM（5 mg· kg^-1· bw ） or equal volume of NS as control, and received intraperitoneal injection of Bai（12.5 mg ·kg^-1· bw） or the same volume of NS as vehicle for 28 d. The hydroxypmline content, the collagen area, the mRNA expression of Col Ⅰ （α）, and the myofibroblasts in lung were examined. Results： The content of hydroxyproline, the percentage of collagen area, the mRNA expression of Col Ⅰ （α）, and the amount of myofibroblast were increased in lungs of rats on day 28 after intratraeheal instillation of BLM, compared with that in lungs of the control rats, respectively（ All P 〈 0.01 ）. The above abnormal changes were ameliorated by Bai （12.5 mg· kg^-1· d^-1 ip, × 28 d （All P 〈 0.05）. Conclusion： Bai has antiaction on BLM-induced fibrosis in lung, and that the above action of Bai is related to the blockage of synthesis of type Ⅰ collagen and the decrease of myofibroblast in lung.
