摘  要:目的通过观察非酒精性脂肪性肝病(NAFLD)大鼠肝脏抵抗素mRNA的表达,探讨其在NAFLD发病机制中的作用。方法24只雄性Wistar大鼠随机分为对照组(普通饮食)和模型组(高脂饮食),各组12只。造模8周、12周末分批处死大鼠,检测血清肿瘤坏死因子-α(TNF-α)、甘油三酯(TG)、谷丙转氨酶(ALT)及内毒素(ET),测定空腹血糖(FBG)、空腹胰岛素(FINS),并计算胰岛素抵抗指数(IRI);应用RT-PCR法检测肝组织抵抗素mRNA的表达;取肝标本做HE染色,观察其病理变化。结果模型组大鼠肝脏抵抗素mRNA的表达量与对照组比较差异有统计学意义(P<0.05),且12周较8周表达量增高。模型组大鼠血清TNF-α、ET、IRI较同期对照组相比均显著升高(P<0.05),12周模型组血清TG、ALT、FBG较同期对照组比较升高(P<0.05)。相关性分析显示,抵抗素mRNA表达量与IRI、TNF-α、ET均呈正相关,TNF-α与IRI和ET亦呈正相关。结论NAFLD大鼠肝脏抵抗素表达量增加,抵抗素可以通过胰岛素抵抗和对炎症因子的调控参与NAFLD的发生与发展。Objective To investigate the expression of resistin mRNA in livers of rats with non-alcoholic fatty liver disease ( NAFLD), and to explore the role of resistin in the pathogenesis of NAFLD. Methods Twenty-four male Wistar rats were randomized into 2 groups:control group (fed with a standard diet)and model group( fed with a high-fat diet). The rats were executed at the end of week 8 and 12, respectively. The serum levels of TG, ALT, TNF-α, FBG, FINS and ET were detected at the end of week 8 and 12, respectively. The IRI was calculated. The expression of resistin mRNA was detected with RT-PCR. The section of liver was fixed in 10% buffered formalin,processed and embedded in paraffin for HE staining. Results The expression of resistin mRNA in livers of rats in model group was significantly higher than that in control group (P 〈 0.05 ). The levels of TG, ALT and FBG in model group were significantly higher than that in control group at the end of week 12( P 〈 0.05 ). The levels of TNF-α, and IRI in model group were significantly higher than that in control group ( P 〈 0.05 ). At the end of week 8 and 12, the level of ET in portal vein in model group increased significantly compared with control group (P 〈 0.05 ). A positive correlation was found between the expression of resistin mRNA and the level of IRI, TNF-α and ET. The level of TNF-α was positively correlated with ET and IRI. Conclusion The expression of resistin mRNA increases in livers of NAFLD model rats. Resistin may participate in the development of NAFLD by insulin resistance and regulating inflammatory cytokines.
