摘  要:目的:探讨CD44v6修饰树突状细胞(DC)融合瘤苗抗胰腺癌的作用,为肿瘤免疫治疗提供实验依据及理论基础。方法:(1)取小鼠骨髓细胞在细胞因子GM-CSF、IL-4、TNF-α的诱导下体外大量扩增DC细胞;(2)在PEG的作用下,进行DC和小鼠胰腺癌细胞(TD2)的融合,并利用HAT/HT系统筛选出纯净融合细胞株;(3)Lipofectamine2000的介导下,将pBud-CD44v6转染到肿瘤融合细胞中表达;(4)荷瘤小鼠接种融合瘤苗,测量肿瘤大小。结果:成功构建了CD44v6修饰的DC/TD2融合瘤苗;免疫组肿瘤体积明显小于对照组。结论:小鼠骨髓来源的DC在PEG作用下成功与TD2细胞融合,同时以CD44v6加以修饰,且融合细胞可有效诱导T淋巴细胞产生强大抗胰腺癌作用。Objective :To study the antitumor effect of fusion hybrid of dendritic eells(DC) and pancreatic cancer cell TD2 expressing CD44v6 for providing the experimental and theoretical foundation on tumor immunotherapy. Methods:(1)Bone marrow-derived mice DCs were largely expanded with cytokine of GM-CSF, IL-4 and TNF-α; (2)DCs and TD2 ceils were fused by polyethylene glyeal(PEG), and the completely fused ones were screened by HAT/HT system ;(3)The recombinant pBud-CD44v6 vector was transfected into the fused cells (hybridoma cell) by lipofectamine 2000, and identhqed by RT-PCR;(4)Fused cells were implanted to bearing--cancer mice, and the volume of tumors were studied. Results:DC/TD2 fused cell vaccine modified with CD44v6 was successfully constructed;the volume of tumors of immune group was smaller than that of control ones. Conclusion:Mice bone-marrow derived DC cells can be fused with TD2 cells by PEG, and the fused ceils can induce strong antitumor immune effect of T lymphocyte.
