摘  要:目的探讨7-二氟亚甲基-5,4’-二甲氧基金雀异黄素(7-difluoromethyl-5,4’-dimethoxygenistein,dFMGEN)拮抗高糖诱导猴视网膜微血管内皮细胞(retinal capillary endothelial cells,RCEC)损伤的浓度效应。方法用不同浓度的dFMGEN和金雀异黄素(genistein,GEN)作用于高糖损伤的猴RCEC。测定各组乳酸脱氢酶活性,MTT法测定细胞增生抑制率,流式细胞术测定细胞凋亡率,观察不同浓度的dFMGEN及GEN拮抗高糖诱导RCEC损伤的浓度效应关系。结果3.0μmol·L-1和10.0μmol·L-1dFMGEN组及GEN组细胞乳酸脱氢酶(LDH)活性分别为(106.600±1.172)U·L-1、(96.900±1.677)U·L-1、(99.400±1.113)U.L-1,细胞增生抑制率分别为29.9%、27.3%和29.1%,细胞凋亡率为(20.20±2.35)%、(9.27±1.30)%、(12.80±4.43)%,与高糖损伤组比较差异均有统计学意义(P均<0.05)。结论dFMGEN能拮抗高糖诱导体外培养的RCEC损伤,并且在一定程度上较其先导化合物GEN所需的浓度低而作用更明显,这种拮抗作用呈明显的浓度依赖性,随浓度的提高作用越明显。Objective To investigate the preventive concentration of 7-difluoromethyl-5,4'-dimethoxygenistein(dFMGEN) on the damage of retinal capillary endothelial cells(RCEC) induced by high glucose. Methods Different concentrations of dFMGEN and GEN were added to monkey RCEC induced by high glucose. The activity of lactic acid dehydrogenase was detected, inhibition ratio for cell proliferation was determined by MTT chromatometry, cell apoptosis ratio was detected with flow cytometry, and effects of different concentrations of dFMGEN and GEN on damage of RCEC induced by high glucose were observed. Results The activity of lactic acid dehydrogenaseof3.0 μmol·L^-1 and 10.0 μmol·L^-1 dFMGEN groups and GEN group were ( 105. 600 ± 1. 172 ) U ·L^-1, (96. 900 ± I. 577 ) U·L^-1, and (99. 400 ± 1. 113 ) U·L^-1, respectively;Inhibition ratios for cell proliferation were 29.9% ,27.3% , and 29. 1% , respectively; Apoptosis ratios for cell proliferation were (20.20 ± 2.35 ) % , (9.27 ± 1.30) % and ( 12.80 ±4.43 )% , respectively. There was significant difference for one of former groups compared with high glucose damage group ( all P 〈 0.05 ). Conclusion dFMGEN can prevent the damage of RCEC induced by high glucose, and it has obvious effect just with low concentration compared with lead compound GEN to some extent. The antagonistic effect is increasing in a dose-dependent way.
