摘 要:目的:探讨表柔比星(epirubicin)对乳腺癌微球体细胞和单层细胞的不同作用。方法:MCF-7细胞在干细胞培养条件下进行微球体培养,MTT法检测表柔比星对MCF-7微球体细胞和单层细胞的抑制率,FCM法检测表柔比星作用下,MCF-7微球体细胞和单层细胞中CD44+CD24-的表达及细胞周期变化。结果:相同质量浓度下,当表柔比星的质量浓度>100ng/mL时,对MCF-7微球体细胞的抑制率明显低于对单层细胞的抑制率(P<0.01);表柔比星(400ng/μL)作用72h后,MCF-7微球体细胞的CD44+CD24-/low比例为(22.8±4.8)%,高于单层细胞的(3.3±0.8)%(P<0.01);MCF-7微球体细胞含有较高比例的G0/G1期细胞(74.33±3.20)%,高于MCF-7单层细胞的(53.40±3.45)%(P<0.01),表柔比星对微球体细胞与单层细胞的G0/G1期影响较小,但显著影响S期及G2期的比例。结论:表柔比星对MCF-7微球体细胞的细胞毒作用较低,并可用于富集乳腺癌干细胞,其对微球体细胞G0/G1比例影响较小。Objective: To explore the different effects of epirubicin on the mammosphere cells and the monolayer cells of breast cancer MCF-7 cells.Methods:MCF-7 cells were cultured in suspension as stem cells to generate primary mammospheres.The inhibitory effects of epirubicin on MCF-7 mammosphere cells and the monolayer cells were measured by MTT assay. The expression of CD44+CD24- and cell cycle distribution in MCF-7 mammosphere cells and the monolayer cells were analyzed by flow cytometry. Results:When the concentration of epirubicin was higher than 100 ng/mL, the cell inhibition ratio was lower in MCF-7 mammosphere cells than that in the monolayer cells at the same concentration of epirubicin (P〈0.01). The CD44+CD24- expression ratio was significantly higher in MCF-7 mammosphere cells than that in the monolayer cells after treatment with 400 ng/μL epirubicin for 72 h, [(22.8±4.8)%] vs [(3.3± 0.8)%] (P〈0.01). The MCF-7 mammosphere cells had higher proportion of cells in G0/G1 phase than the monolayer cells, [(74.33±3.20)%]vs[(53.40±3.45)%] (P〈0.01). Epirubicin had little effect on the cells in G0/G1 phase of MCF-7 mammosphere cells and the monolayer cells, but had a great influence on the cell proportion in S and G2 phase. Conclusion: Epirubicin had lower cytotoxicity on MCF-7 mammosphere cells and it can be used to enrich breast cancer stem cells. Epirubicin had little effect on the cell proportion in G0/G1 phase of MCF-7 mammosphere cells as compared with control.
