摘  要:目的探讨喉癌中PTTG、bFGF的表达以及在新生血管生成过程中的作用。方法应用免疫组化SP法分别检测57例喉癌、26例癌旁和9例正常喉黏膜中PTTG、bFGF蛋白的表达,用CD34标记并计数肿瘤内微血管密度。结果喉癌中PTTG蛋白和bFGF蛋白表达率分别为84.2%和78.9%,明显高于癌旁及正常喉黏膜的表达率(P<0.05)。PTTG的表达与肿瘤分化程度、临床分期相关和淋巴结转移差异有统计学意义(P<0.05),bFGF的表达与肿瘤分化程度和淋巴结转移差异有统计学意义(P<0.05),与临床分期差异无统计学意义(P>0.05)。两者的表达均与患者年龄、性别和肿瘤原发部位差异无统计学意义(P>0.05),且两者之间呈显著正相关(P<0.05)。喉癌中MVD值明显大于癌旁和正常喉黏膜(P<0.05),并与淋巴结转移显著相关(P<0.05),MVD分别与PTTG和bF-GF的表达相关(P<0.05)。结论PTTG和bFGF的表达与喉癌的临床病理学特征密切相关,二者的检测将有助于喉癌的诊断、评估喉癌患者的恶性程度。Objective To explore expressions of the pituitary tumor-transforming gene(PTTG) and basic fibroblast growth factor (bFGF) proteins and their relationship to MVD in laryngeal carcinoma. Methods Expressions of PTTG and bFGF were determined by the immunohistochemical SP method in 57 cases of laryngeal carcinoma, and compared with those in 26 cases of paracarcinoma tissues and 9 cases of normal tissues MVD in laryngeal carcinoma was evaluated by immunohistochemical staining with antibody CD34. Results The positive rate of PTTG and bFGF was 84.2 % and 78.9% in laryngeal carcinoma, which was significant higher than those in para-carcinoma tissues and nonnal tissues( P 〈 0.05). Expression of PTTG was associated with histological differentiation and clinical stages( P 〈 0.05) and was significantly associated with lymph node metastases( P 〈 0.05), but was not associated with age, gender and primary lesions( P 〉 0.05). Expression of bFGF was significantly associated with histological differentiation and lymph node metastases( P 〈 0.05), but was not associated with age, gender, clinical stages and primary lesions( P 〉 0.05). There was a significant correlation between expression of PTTG and bFGF( P 〈 0.05). The value of MVD in laryngeal carcinoma was significantly higher than that in para-carcinoma tissues and normal tissues( P 〈 0.05), and was significantly associated with lymph node metastases( P 〈 0.05), but not associated with age, gender, histological differentiation, clinical stages and primary lesions( P 〉 0.05). Expressions of PTTG and bFGF were correlated with MVD in laryngeal carcinoma ( P 〈 0.05). Conclusion Increased PTTG may play an important role in the carcinogenesis and development in laryngeal cancers by promoting expression of bFGF protein and angiogenesis.
