摘 要:目的:观察不同浓度尼古丁对体外培养的人牙周膜细胞(periodontal ligament cells,PDLCs)增殖及合成纤维结合蛋白(fibronectin,Fn)的影响,探讨吸烟对牙周炎的影响机制。方法:不同浓度的尼古丁作用于PDLCs,分别用MTT比色法、酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)和逆转录-聚合酶链式反应(RT-PCR)检测尼古丁对PDLCs增殖及合成Fn的影响。结果:①MTT法:尼古丁能明显抑制牙周膜细胞的增殖(P<0.05),1μg/ml尼古丁显示出最佳的抑制增殖作用(P<0.01);②ELISA法:浓度为50ng/ml~3μg/ml的尼古丁抑制PDLCs合成Fn(P<0.05),且呈浓度依赖性;③RT-PCR法:从分子生物学水平进一步证明尼古丁抑制Fn的合成。结论:尼古丁抑制牙周膜细胞的增殖及Fn的合成,并呈浓度依赖性,说明一定浓度的尼古丁影响牙周膜细胞的增殖及在根面的附着,加重牙周组织炎症,进一步影响牙周新附着的形成。Objective:To observe the effects of nicotine on the proliferation and fibronectin (Fn) synthesis of human periodontal ligament cells(PDLCs) cultured in vitro, and to investigate the mechanism of the effects of smoking on the periodontitis, nethods:PDLCs were cultured in the presence of nicotine at various concentrations, the proliferation of cells was measured by MTT chromatometry, and the synthesis of Fn was measured by ELISA and reverse transcription- polymerase chain reaction ( RT- PCR ). Results : ① MTT chromatometry : Nicotine inhibited the proliferation of PDLCs obviously ( P 〈 0.05 ) , and there was optimal effect when the concentration of nicotine reached to 1 μg/ml( P 〈 0.01 ). ②ELISA : The synthesis of Fn decreased when the concentration of nicotine increased from 50 ng/ml to 3 μg/ml ( P 〈 0.05 ). ③RT- PCR : It demonstrated that nicotine inhibited the synthesis of Fn mRNA. Conclusion: Nicotine can inhibit the proliferation and the synthesis of Fn of PDLCs with concentration-dependence. It suggests that nicotine with definite concentration affected the proliferation and the attachment to the teeth root. Nicotine can aggravate the periodontitis and affect the formation of new-attachment of periodontium.
