摘  要:目的研究不同剂量、不同的用药顺序等配伍方式,探讨干扰素与化疗药物体外联合应用,对人胃癌细胞株的抑制作用.方法以人胃癌细胞株FGC85作为靶细胞,用肿瘤细胞集落形成法进行药敏实验.分析了干扰素α2b(IFNα2b)与氟脲嘧啶、顺铂联合应用时,不同的配伍方式对人胃癌细胞株增殖的抑制作用.实验分组为:①化疗药物;②IFNα2b;③化疗药物和IFNα2b同时作用;④化疗药物继以IFNα2b;⑤IFNα2b继以化疗药物.结果IFNα2b与氟脲嘧啶或顺铂联合应用对FGC85细胞增殖的抑制作用明显强于各单独应用,各组抑制率之间均有显著差异(P<001);IFNα2b降低氟脲嘧啶、顺铂的IC50,并且有显著差异(P<005,P<001);加药顺序不同,抑制效果亦不同,以化疗药物继以IFNα2b组最佳,IFNα2b继以药物组最差,二者间有显著差异(P<001).结论IFNα2b与氟脲嘧啶、顺铂联合应用,可提高药物的抑瘤作用.AIM To study the inhibitive effect of combined IFN and chemotherapeutic drugs at different doses and regimens in human gastric cancer cell line in vitro . METHODS The drug sensitivity on human gastric cancer cell line FGC 85 was determined by colony formingmethod. The synergistic effects of IFNα 2b, 5 FU and DDP in different schemes on FGC 85 were analyzed. Five groups were used: ①adding chemotherapeutic drugs alone, ②adding IFNα 2b alone, ③adding chemotherapeutic drugs and IFNα 2b simultaneously, ④adding chemotherapeutic drugs following IFNα 2b, and ⑤adding IFNα 2b following chemotherapeutic drugs. RESULTS When 5 FU or DDP was combined with IFNα 2b on FGC 85 cell line, the inhibition turned to be more pronounced as compared with 5 FU or DDP alone. The inhibition rate was significantly different from each other ( P <0 01). The IC 50 of 5 FU or DDP was significantly lower when IFNα 2b was added ( P <0 05 and P <0 01). The inhibitive effects were significantly different with different orders of adding drugs and IFN. The effects of adding chemotherapeutic drugs following IFNα 2b were significantly better than that of adding IFNα 2b after chemotherapeutic drugs ( P <0 01). CONCLUSION The inhibition of tumors was more pronounced when IFN and 5 FU or DDP were given in combination.
