摘 要:目的:探讨光卟啉(Photofrin)不同孵育浓度和不同光照能量密度对人食管癌细胞Eca-109体外光动力治疗(photodynamic therapy,PDT)效应的影响。方法:以不同浓度Photofrin孵育Eca-109细胞,并在不同光能量密度(10、30、50J/cm2,光源采用DIOMED630PDT系统)下行PDT,24h后通过MTT法检测细胞生存率。结果:Photofrin三种不同光照能量密度及不同浓度下Eca-109细胞生存率间差异均有显著性(均P<0.01)。同一光照能量密度下不同浓度间其生存率差异均有显著性(均P<0.01),而同一浓度下不同光照能量密度间的生存率,则除了浓度为10.0μg/mL间无明显差异外,其余差异均有显著性(均P<0.05)。孵育浓度和光照能量密度间存在显著交互效应(P<0.01)。结论:特定光源下,Photofrin的孵育浓度和光照能量密度对人食管癌细胞Eca-109体外PDT效应影响显著。Objective To investigate the influence of photofrin photodynamic therapy effect with different incubating concentration and light energy density in human esophageal cancer cell line in vitro. Methods Human esophageal cancer cells were incubated in vitro with photofrin in different concentration incubation and different light energy density (10, 30, 50 J/cm^2), and then irradiated in DIOMED630PDT systems, survival rate of each concentration was measured by MTT assay after 24 hours' incubation. Results The cell survival rate between different photofrin incubating concentration and three different light energy density and under the same energy density with different concentration are all significant different (all P 〈 0.01 ). While under the same concentration, the cell survival rate is also significant different with different light energy density except that the concentration is 10.0μg/mL (all P 〈 0.05). And there is significant interaction between incubating concentration and light energy density (P 〈 0.01).Conclusion Under the special light source, the photofrin incubating concentration and light energy density were significant influence to photodynamic therapy effect in human esophageal cancer cell line in vitro.
