摘 要:通过逆转录-聚合酶链反应从我国河南省2例重叠感染HCV或HBV/HDV的献血员中,分离到HGVNS5区的部分cDNA,对其进行序列分析比较,结果表明:河南株HGVNS5区核苷酸与两株中国HGV株的同源性(>91.9%)高于国外代表株(88.5%-90.6%),但由核苷酸推导的氨基酸的同源性都无明显的地区性区别。HGVNS5区氨基酸序列较保守(同源性>91%),缺乏明显高变区。中国4株HGV在7384位发生了由C→T的变异,从而导致一个共同的保守位点的产生(由F→A)。重叠感染其他肝炎病毒时。Non-structural region 5(NS5)gene fragments were isolated from two blood donors in Henan Province who were co-infected with hepatitis C or B/D viruses.These fragments were obtained by using reverse transcription and polymerase chain reaction(RT-PCR),and these overlapping fragments were cloned into pGEM-T vector.Sequences were determined and then analyzed by computer.The results indicated as follows:the nucleotide identity of HGV NS5 cDNA among the Henan isolates and the other Chinese isolates(HGVCN,HGVC964)were higher(>91.9%)than that among Henan and foreign country(Africa,American,Japan,Europe)isolates(88.5%-90.6%);but the amino acid homology didn’t show any significant difference among them.The NS5 fragments of HGV strains compared were very conserve(above 91.9% identical to each other)in amino acid level and didn’t exist distinct hypervariable region.Four Chinese isolates of HGV consisted a common variation(C→T)in 7384 nucleotide (the position number according to U44402)and this led to a unique amino acid variation(F→A).It can be suggested that the HGV fragments isolated from Henan area didn’t exist characteristic change when they were co-infected with other hepatitis viruses.
