摘  要:用MTS比色法测定了不同质量浓度的板蓝根多糖对体外培养的猪脾脏淋巴细胞增殖的影响,分别用ELISA法和硝酸还原酶法测定了板蓝根多糖对猪脾脏淋巴细胞分泌IL-2,IL-4,IFN-γ和NO的影响.结果表明,板蓝根多糖能显著促进猪脾脏淋巴细胞的增殖,同时又能协同ConA或LPS诱导的猪脾脏淋巴细胞增殖(P<0.05);能显著促进由ConA诱导的猪脾脏淋巴细胞分泌IFN-γ,抑制IL-2的分泌,对IL-4分泌的影响不明显;低剂量板蓝根多糖能显著促进NO的分泌(P<0.01).The study was conducted to investigate the mechanism of immuno-regulation induced by isatis root polysaccharide (IRPS). The effects of the IRPS on proliferation of swine splenic lymphocytes in vitro was tested by the method of MTS colorimetric assay. NO was detected by reduction of nitrate method and IL-2, IL-4,IFN-γ were detected by ELISA. The results showed that IRPS could stimulate proliferation of swine splenic lymphocytes induced by ConA or LPS significantly ( P 〈 0.05 ) in vitro. Upregulation of IFN-γ co-induced by ConA and IRPS in all three dose groups were observed. In contrast, IRPS appeared to inhibit the secretion of IL-2 induced by ConA and had no influence on IL-4 induced by ConA . The level of NO secreted by swine splenic lymphocytes was enhanced significantly (P 〈 0.05 )by IRPS in the low dose group.
