摘 要:目的探讨Smac和Livin在肾细胞癌(肾癌)组织中的表达及两者在肾癌细胞凋亡中的作用和关系。方法应用半定量RT-PCR技术检测49例肾癌组织和20例正常肾组织Smac和Livin mRNA表达,酶联免疫吸附试验(ELISA)检测Smac和Livin蛋白表达情况。结果Smac mRNA及其蛋白在肾癌组织中的表达显著低于正常肾组织(χ2=5.929、6.538,P<0.05),Livin mRNA及其蛋白在肾癌组织中的表达显著高于正常肾组织(χ2=27.600、23.351,P<0.05),且两者均与肾癌临床病理参数有关。在肾癌组织中,Smac和Livin表达强度之间呈负相关(r=-0.39,P<0.05)。结论Smac在肾癌组织中低表达,其表达与Livin呈明显的负相关,两者可能是肾癌细胞凋亡信号传导网络中的重要一环。Objective To investigate the expressions of Smac and Livin and their association with apoptosis in renal cell carcinoma (RCC). Methods The expressions of Livin and Smac mRNA were detected using reverse transcription-polymerase chain reaction (RT-PCR) and ELISA for tissues of 49 RCC and 20 normal kidneys. Results Expressions of Smac mRNA and protein in RCC tissue were significantly lower than that in normal renal tissue (x^2=5. 929,6. 538;P〈0.05), while expressions of Livin mRNA and protein in RCC tissue were higher than the normal one (x^2=27. 600,23. 351 ; P〈0.05). Both Smac and Livin expressions were related with parameters of clinical pathology. A negative correlation was observed between the expressions of Smac and Livin in RCC (r=-0.39,P〈0.05). Conclusion Smac expression was lower in RCC, which shows a negative correlation with Livin. These two genes are probably an important link in the apoptosis signal transduction network of RCC cells.
