摘  要:目的探讨脂肪酸合酶(FAS)抑制剂浅蓝菌素能否阻遏人骨肉瘤细胞增殖与诱发凋亡。方法采用人骨肉瘤细胞系U2-OS,在体外实验中应用噻唑蓝法(MTT法)观察浅蓝菌素对人骨肉瘤细胞U2-OS生长的抑制作用,并通过流式细胞仪对细胞凋亡和细胞周期变化进行检测。结果U2-OS细胞在浅蓝菌素作用下,细胞增殖被阻遏,并呈现剂量效应关系,2.5、5.0、10.0和20.0mg/L浅蓝菌素作用24h的抑制率分别为(18.58±2.49)%、(35.11±3.49)%、(56.60±3.53)%和(79.69±2.83)%,与对照组比较有显著性差异(P<0.05)。流式细胞仪显示浅蓝菌素可诱发骨肉瘤细胞发生凋亡并且此作用呈剂量、时间依赖趋势;细胞周期分析浅蓝菌素能阻滞肿瘤细胞从S期进入G2/M期,并诱导其细胞凋亡。结论浅蓝菌素抑制FAS可阻遏骨肉瘤细胞增殖,其机制可能是诱导细胞发生S期阻滞和细胞凋亡。Objective To investigate whether Cerulenin could induce apoptosis and suppress the proliferation of human osteosarcoma cells. Methods The inhibitory effect of FAS inhibitor Cerulenin on the growth of human osteosarcoma cells(U2-OS) was determined by tetrazolium assay in vitro. Flow cytometry (FCM) analysis was used to study the cell apoptosis and changes of cell cycle. Results Cerulenin was able to induce U2-OS cells death via apoptosis. Cerulenin also inhibited the proliferation of cells in vitro based on dose-dependence. The inhibition rates of U2-OS cells after 24 hours exposure to Cerulenin at 2.5,5. o, 10.0 and 20.0 mg/L were respectively (18.58±2.49)%,(35.11±3.49)%,(56. 60±3.53)% and (79.69±2.83)% (P〈0.05 vs controls). FCM analysis showed that after treatment with different concentrations of Cerulenin, the apoptosis rate of human osteosarcoma cells significantly increased in a dose and time dependent manner. Cerulenin arrested the transition of U2-OS cells from S to G2/M phase,and induced cellular apoptosis. Conclusion Its suggests that Cerulenin could inhibit proliferation of U2-OS cells in that it induces apoptosis and S phase arrest.
