摘 要:目的:探讨咖啡酸苯乙酯(Caffeic acid phenethyl ester,CAPE)在人神经胶质瘤中的治疗作用。方法:建立裸鼠胶质瘤模型,观察CAPE在体内对肿瘤的影响。运用实时荧光定量PCR、Westernblotting检测CAPE对胶质瘤细胞株U251异常Wnt/β-catenin通路及靶基因c-myc的影响。结果:通过对荷瘤鼠模型的观察,发现口服CAPE可以抑制神经胶质瘤的生长。CAPE可以抑制β-catenin基因mRNA及蛋白的表达,下游靶基因c-myc的mRNA及蛋白表达也相应降低。结论:CAPE可以抑制神经胶质瘤异常Wnt/β-catenin通路中β-catenin基因及c-myc基因的表达,对神经胶质瘤的生长有抑制作用。Objective: The therapic effect of caffeic acid phenethyl ester ( CAPE ) in neuroglioma was to be investigated in this article. Methods: Athymic mouse model was constructed for therapic observation when tumor was treated by CAPE in vivo. Then Real-time PCR was employed to detected the expressing level of c-myc and β-catenin genes in Wnt/β-eatenin signaling pathway when U251 cells were treated by CAPE. Results: CAPE could inhibit the growth of neuroglioma in athymic mouse through drug oral application. The mRNA and protein expression of β-catenin were inhibited. Furthermore, c-myc gene, which was the downstream of β-catenin gene, was also down-regulated after the drug treatment. Conclusion: The expressing level of β-catenin and e-mye genes were down-regulated after CAPE treatment, by which the growth of neuroglioma could be inhibited.
