摘 要:目的探讨肾炎四味片对单侧输尿管梗阻大鼠肾间质纤维化的影响及其作用机制。方法采用单侧输尿管结扎(UUO)肾间质纤维化模型,将SD大鼠60只随机分为4组,即假手术组、肾炎四味片假手术组、模型组和肾炎四味片治疗组,每组15只,分别于术后第3、7、14 d每组大鼠各取5只。用HE和Masson染色法观察肾脏病理改变,用免疫组化方法检测肾小管间质中转化生长因子β1(TGF-β1)和血小板凝血酶敏感蛋白-1(TSP-1)表达情况,对结果进行半定量分析。结果假手术组和肾炎四味片假手术组肾小管间质胶原纤维相对面积及TGF-β1、TSP-1的表达组内及组间比较无统计学意义(P>0.05)。模型组肾小管间质胶原纤维相对面积及TGF-β1、TSP-1的表达,随梗阻时间延长而增加(P<0.05),且与同期假手术组比较差异有统计学意义(P<0.05);在术后各时间点,肾炎四味片治疗组肾小管间质胶原纤维相对面积及TGF-β1、TSP-1的表达较模型组显著减少(P<0.05),但仍高于假手术组(P<0.05)。结论肾炎四味片缓解UUO大鼠肾间质纤维化可能部分是通过降低肾脏TGF-β1和TSP-1的表达而实现的。Objective To explore the effects and mechanism of Shenyansiwei Tablet on renal interstitial fibrosis in unilateral ureteral obstruction rats. Methods Rats model with renal interstitial fibrosis were established by unilateral ureteral obstruction of (UUO). Sixty male SpragueDawley rats were randomly divided into sham operation (group A), shenyansiwei Tablet sham operation (group B), unilateral ureteral obstruction (group C) and shenyansiwei Tablet treatment groups (group D). The animals were sacrificed at 3, 7 and 14 days after operation separately. The histological changes of renal interstitial tissues were observed by HE and Masson staining. The sites and levels of TGF - β1 and TSP - 1 were detected by immunohistochemistry staining. Results There was no significant difference in the same group at the three different time points and between the two groups of the same stage about group A and B. The renal histological changes in group C gradually aggravated, there was significant difference at the three different time points( P 〈 0.05) ; the renal histological changes in group D decreased compared with group C( P 〈0.05). Compared with group A, the expression of TGF- β1 and TSP- 1 in the renal interstitum of group C increased significantly ( P 〈 0.05). After treatment with Shenyansiwei Tablet, the ex- pression of TGF - β1 and TSP - 1 was decreased ( P 〈0. 05). Conclusions Regulation of expression of TSP - 1 and TGF - β1 may be responsible for preventing progression of renal interstitial fibrosis of Shenyansiwei Tablet in UUO rats.
