摘  要:目的研究两种端粒酶抑制剂干扰素α2b(IFN—α2b)和顺铂(CDDP)对人葡萄膜黑色素瘤细胞生长抑制作用,为临床化疗人葡萄膜黑色素瘤提供依据。方法用不同浓度及不同作用时间的IFN-α2b和CDDP作用于体外培养的人葡萄膜黑色素瘤细胞。倒置显微镜、电子显微镜观察其形态学变化,并用MTT法检测干扰剂对细胞生长的抑制作用。结果随着IFN—α2b和CDDP浓度的增加及作用时间的延长,细胞出现生长抑制的形态学变化,电子显微镜显示细胞出现凋亡。作用72h,IFN-α2b和CDDP浓度分别大于500IU/ml及1.00mg/L时细胞出现抑制状态,并在浓度分别达到5000IU/ml及10.00mg/L时,其细胞抑制率(66.1±4.7)%及(74.0±3.3)%与对照组差异有统计学意义(P〈0.001)。IFN—α2b(5000IU/ml)和CDDP(10.00mg/L)浓度固定,作用时间达到48h后,细胞出现抑制状态;72h后抑制率(67.9±8.1)%及(76.5±1.6)%与对照组差异有统计学意义(P〈0.001);即随着两者浓度升高及作用时间增长,葡萄膜黑色素瘤细胞受到的抑制作用逐渐增强(P〈0.01)。结论端粒酶抑制剂IFN-α2b和CDDP可有效抑制体外培养的人葡萄膜黑色素瘤细胞的生长,且随着两者浓度升高及作用时间延长抑制作用加强,并导致细胞凋亡。Objective To study the inhibitory effects of two telomerase inhibitors, IFN-α 2b (interferon-ct2b) and CDDP (eisplatin, cis-dichlorodiamine platinum), on uveal melanoma cells of hmnan eyes, and to provide evidence for chemotherary of uveal melanoma. Methods IFN- α 2b and CDDP of different concentration and duration were cultured with uveal melanoma cells of human eyes , and compared with control group. Morphological changes of cells were observed by inverted optical and electronic microscope, and the restrain effects were detected by MTT assay. Results More and more obvious morphological changes of restraint and apoptosis of cells were observed with the increasing concentration and duration of IFN- α 2b and CDDP. When action duration was 72 h, the cells were inhibited with the concentration more than 500 IU/ml (IFN-α2b) and 1.00 mg/L (CDDP) respectively. The cells restrain rates were (66.1 ±4,7)% and (74.0±3.3)% with the concentration more than 5000 IU/ml (IFN-α2b) and l0 mg/L (CDDP), and the rates were higher than control (P〈0.01). When concentrations were 5000 IU/ml (IFN-oL2b) and 10.00 mg/L (CDDP), cells were inhibited at 48 h and the restrain rates were (67.9±8.1)% and (76.5± 1.6)% at 72 h, respectively, (P〈0.01). With action duration or concentration of telomerase inhibitor increasing, the restrain effect on uveal melanoma cells of human eyes gradually increased (P〈0.01).Conclusionss As two effective telomerase inhibitors, IFN-ct2b and CDDP can effectively restrain the growth of uveal melanoma cells. The restrain effect gradually increases with action duration or concentration of telomerase inhibitor increasing, which can cause cultured cells apoptosis.
