摘  要:目的探讨热休克蛋白70(TB.HSP70)-H22肽是否能促进树突状细胞(DC)活化成熟并诱导H22特异性抗肿瘤免疫。方法取小鼠骨髓于培养第7天分为A、B、C 3组,分别给予未刺激、TB.HSP70-H22肽(10μg/mL)、TB.HSP70-H22肽(20μg/mL)干预24h,取上清液用ELISA法检测IL-12、IL-10,用FACS法检测CD40、CD80,用混合淋巴细胞反应检测淋巴细胞增殖,用MTT法检测DC活化淋巴细胞对H22和B16肿瘤细胞杀伤率。结果与A组比较,B、C组除对B16细胞杀伤率差异无统计学意义外,B、C组IL-10明显降低,CD40、CD80、IL-12、淋巴细胞增殖程度和对H22杀伤率均明显升高,差异有统计学意义(P<0.05);与B组比较,C组对B16细胞杀伤率差异无统计学意义,IL-10明显降低,CD40、CD80、IL-12、淋巴细胞增殖程度和对H22杀伤率均明显升高,差异有统计学意义(P<0.05)。结论TB.HSP70-H22肽能明显促进DC活化成熟,诱导H22特异性抗肿瘤免疫,为该疫苗用于肝癌免疫治疗提供了理论基础。Objective To study whether TB. HSP70 loaded with H22-peptide from Pichia pastor could facilitate activation and maturity of dendritic cells(DCs) ,and induce specific anti hepatoma immunity. Methods The murine marrow cells were cultured for 7 days,then randomized to 3 groups(A, B, C), and stimulated respectively for 24h by no stimulus, Hsp70-H22 peptide complexes (10μg/mL) and Hsp70 H22 peptide complexes(20μg/mL). IL-12 and IL-10 of supernatant were tested by ELISA assay,CD40 and CD80 of DCs were test by FACS and the proliferation of spleen lymphocytes were test by mixed lymphocyte reaction. The ability of spleen lymphocytes activated by DCs to H22 and B16 cells in vitro was investigated by MTT. Results Compared with A group,ex- cept for the B16 tumor cell-killing effect, the level of IL-10 of group B and C remarkably decreased, the levels of CD40,CD80,IL-12, the proliferation of spleen lymphocytes and the H22 tumor cell-killing effect remarkably increased,these differences had statistical significance(P〈0. 05). Compared with B group,except for the B16 tumor cell-killing effect,the level of IL-10 of group C remark- ably decreased, the level of CD40,CD80, IL-12, the proliferation of spleen lymphocytes and the H22 tumor cell-killing effect remark- ably increased, these differences had statistical significance (P〈 0. 05). Conclusion TB. HSPT0 loaded with H22 peptide from Pichia pastor could remarkably facilitate activation and maturity of dendritic cells(DCs) and induce specific anti-hepatoma immuni ty,and provide rationale for the immunotherapy of hepatoma.
