摘 要:目的:观察小鼠→大鼠异种心脏移植后延迟性排斥反应(DXR)的病理特征,研究E-selectin在异种移植心脏的表达变化,探讨DXR发生机制。方法:袖套法建立小鼠→大鼠颈部异位异种心脏移植模型,将接受异种心脏移植的大鼠随机分为5组,1组(n=16)在供心停跳时取心,余4组(每组n=4)分别在移植术后3、8、16、24 h供心仍跳动时取心,另取未移植小鼠心脏作为对照组(n=4)。各组标本作HE染色、免疫组化SABC法检测E-selectin表达变化。结果:移植心脏HE染色表现为血管内皮损伤、血栓形成、心肌实质损伤、间质出血、炎症细胞浸润等病理特征随移植术后时间逐渐加重,完全排斥时部分心肌凝固性坏死及溶解、广泛间质出血、血管结构崩解及血管内血栓形成、严重炎症细胞浸润。对照组心脏无E-selectin表达,移植组心脏在移植后3 h即有E-selectin表达于血管内皮细胞,且随时间推移表达逐渐增强,排斥时达到高峰。结论:血管内皮细胞激活参与DXR发生,E-selectin表达与DXR发生和发展密切相关,E-selectin表达可早期提示DXR发生。Objective:To study the pathologic features and the expression of E-selectin in delayed cardiac xenograft rejection. Methods:A model of mouse-to-rat cardiac heterotopic xenotransplantation in neck was established by cuff technique. The cardiac xenografts were harvested at 3 h (n= 4), 8 h (n = 4), 16 h (n= 4) and 24 h(n=4) after transplantation while the hearts were still beating. Other cardiac hearts(n= 16) were not harvested until rejection time. Untransplanted mouse hearts were studied as controls (n= 4). All samples were examined by HE and E-selectin expression by immunohistochemistry. Results : During the period of post- transplantation, the degree of rejection in cardiac xenografts became serious gradually till they were ultimately rejected. Immunohistochemical examination showed that E-selectin expression was found as early as 3 h posttransplantation and increased gradually on vascular endothelial cells of xenografts ,whereas the contral group did ont express. Conclusion: E-selectin expression is related with the development of the delayed xenograft rejection ,and endothelial cells activation involves in also.
