摘 要:采用乙酸乙酯作溶剂对仙草中的叶绿素进行浸提,并经皂化、酸化、锌代及成盐反应制备叶绿素锌钠盐.采用分光光度法对叶绿素锌钠盐进行定性、定量测定,确定最大波长为410 nm.通过正交实验及单因素实验确定较佳工艺条件,并对其稳定性进行研究.结果表明:最佳工艺条件为皂化pH=11,酸化pH=2,成盐pH=11,锌代温度65℃,锌代时间1.5 h,硫酸锌用量30 ml,产品得率可达0.46%,而且制备得到的仙草叶绿素锌钠盐具有良好的光稳定性和热稳定性.Chlorophyll, extracted by ethyl acetate from Hsian -tsao( mesona procumbens hemsl), was made into sodium zinc chlorophyllin through saponification, acidification, ironing and basification. The quality and quantity were determined by spectrophotometer, and its maximum absorption wavelength was determined at 410nm. The optimal reaction condition was determined by orthogonal and single factor experiments, and the stability was also studied. The optimum of preparation process was: pH of saponification = 11, pH of acidification = 2, pH of basification = 11, ironing under 65 ℃ with 30 ml ZnSO4 for 1.5 hours. The yield could reach 0.46%. And the product has good stabilities for light and heat.
