摘  要:目的:研究乳腺癌MCF-7细胞系中SP细胞与非SP细胞表面标记的特点,并进行比较分析。方法:以体外培养的MCF-7细胞作为研究对象,采用Hoechst33342染色,并用流式细胞仪分选出SP细胞和非SP细胞,检测两种细胞的各种表面蛋白或抗原和细胞周期增殖状况。结果:乳腺癌MCF-7细胞株中SP细胞的含量为(10.1±0.84)%;分选后SP细胞的CD38、CD117和CD34表达率分别为(1.67±0.60)%、(0.43±0.22)%和(0.27±0.12)%,非SP细胞分别为(1.37±0.49)%、(0.42±0.24)%和(0.33±0.15)%,差异无统计学意义,P均>0.05;两者CD133、CD24、ABCG2的表达率分别为(34.33±7.03)%、(46.68±4.44)%、(85.82±2.48)%和(5.62±1.16)%、(75.38±5.14)%、(4.89±1.80)%,差异有统计学意义,P均<0.05;两者细胞中G0/G1、S、G2/M期细胞数分别为(81.34±6.69)%、(13.48±5.90)%、(5.18±2.42)%和(57.71±6.46)%、(23.14±6.46)%,(19.14±4.61)%,差异有统计学意义,P均<0.05;两者Ki-67表达率分别为(12.63±2.10)%、(34.37±4.71)%,差异有统计学意义,P=0.000。结论:乳腺癌MCF-7细胞系中含有比例较高的SP细胞,且SP与非SP细胞有可以分别鉴定和纯化的抗原差别。选择SP方法分离乳腺癌干细胞具有科学性。OBJECTIVE: To investigate the markers between side population ceils and non-side population cells from MCF-7 cancer cell line. METHODS: After staining with Hoechst33342 dye, side population ceils and non-side population cells were sorted by FACS from MCF-7, and these cell markers, antigens and cycles were measured. RESULTS: The side population ceils containing (10. 1 ± 0.84)% existed in MCF-7. The proportions of CD38, CDl17, CD34 were (1.67 ± 0.60)%, (0.43±0.22)%, (0.27±0.12)% and (1.37± 0.49)%, (0.42±0.24)%, (0. 33±0.15)% in side population cells and non-side population cells, respectively. There was no marked difference (P〉 0.05). The proportions of CD133,CD24, ABCG2 were (34.33± 7.03)% , (46.68 ± 4.44)%, (85.82±2.48)% and (5. 62±1.16)%, (75.38± 5.14) %, (4.89 ± 1.80) % in side population cells and nonside population cells, respectively. There was significant difference (P〈0.05). The proportions of G0/G1 ,S, G2/M, Ki-67 were (81.34±6.69)%, (13. 48 ± 5. 90) % , (5.18± 2.42)%, (12.63±2.10)% and (57.71±6.46)%, (23.14±6.46)%,(19.14±4.61)% , (34. 37±4. 71) % in side population cells and non-side population cells, respectively. There was significant difference (P =0. 000). CONCLUSIONS: Side population cells express high level in breast cancer cell line, and SP and non-SP cells can be identified and purified by using their antigenic markers properties. SP may be a scientific method to isolate breast cancer stem cells.
