含HN和VP3及IL-18重组鸡痘病毒体内抑制骨肉瘤细胞S-180效应的观察

摘  要:目的:研究含凋亡素基因、鸡新城疫病毒的HN基因及IL-18基因的重组鸡痘病毒对小鼠骨肉瘤的抑制效应。方法:建立BALB/C小鼠荷S-180骨肉瘤模型,瘤内注射重组鸡痘病毒,测定肿瘤生长率、抑瘤率和小鼠血清唾液酸含量,肿瘤组织病理学分析检测重组鸡痘病毒对骨肉瘤的抑制效应。结果:rFPVHN、rFPVVP3和rFPV3(rFPVVP3IL-18)均对骨肉瘤有抑制作用,但rFPV3组抑瘤率高于rFPVHN组、rFPVVP3组和鸡痘病毒野毒对照组达到51.7%,血清唾液酸含量明显减低(4.22±0.27),P=0.008 7。结论:凋亡素基因、鸡新城疫病毒的HN基因及IL-18基因联合应用具有协同抑制骨肉瘤的效应。OBJECTIVE: To explore the effective method of tumor gene therapy and studying the antitumor effect of HN gene, VP3 gene and IL-18 gene. METHODS: The osteosareoma model in BALB/c mice was constructed and the rFPV were injected into the tumor. The therapeutic effect of rFPV to BALB/e mice bearing S- 180 tumor was observed, and the rate of tumor growth, rate of tumor suppression, sialic-acid in serum, and pathology analysis were detected by immunohistochemistry. RESULTS: Compared with the control groups (rFPVHN group and rFPVVP3 group), the tumors in the group of rFPV3 (rFPVVP3 IL -18) were decreased obviously, and the rate of tumor suppression reached 51.7 %. The sialic-acid in serum obviously decreased (4. 22 ± 0.27 ), P = 0. 008 7. CONCLUSIONS: The rFPV3 is able to suppress the proliferation of the tumor cells in BALB/ c mice bearing S-180 tumor. HN gene, VP3gene and IL-18 gene have synergistic inhibition effect on osteosareoma.
