摘 要:为探讨兰州市大气可吸入颗粒物(PM10)的生物活性来源,使用质粒DNA评价法和电感耦合等离子质谱(ICP-MS)分别研究了兰州市市区和郊区不同季节PM10全样和水溶部分的生物活性和微量元素浓度.研究结果表明,兰州市不同季节、不同采样点PM10的生物活性差异较大,PM10全样和水溶部分的TD20最低值分别为10μg.mL-1和43μg.mL-1,最高值分别为980μg.mL-1和1150μg.mL-1.ICP-MS分析结果显示,PM10中全样和水溶性Zn元素的平均浓度最高,其次是Pb和Mn元素,且PM10中Zn、Cd和As等元素多以可溶状态存在,而Fe、Pb和V等元素则相反.PM10的TD20值与V、Cr、Mn、Co、Ni、Cu、Zn、As、Cd、Hg、Cs和Pb元素总浓度之间的相关性分析结果表明,兰州市PM10的生物活性主要来自水溶性微量元素.PM10全样和水溶部分的TD20值与单个水溶性Zn、Mn、Pb、Fe、V、Cu、Cr、Cd、Cs和Co元素之间具有显著的负相关性,表明这些水溶性元素可能是导致兰州市质粒DNA损伤的主要原因.Abstract: To investigate the sources of bioreactivity of PM10 collected from urban and suburban sites of Lanzhou in different seasons, an in vitro plasmid DNA assay was employed to study the bioreactivity of intact whole solution and the water-soluble fraction of PM10. ICP-MS was used to determine the trace element concentrations in the solutions. The results showed that the bioreactivity of PM10 collected at two sampling sites in different seasons varied greatly. The lowest TD20 values (toxic dosages of PM10 causing 20% of plasmid DNA damage) of the whole solution and water-soluble fraction of PM10can be as low as 10 μg.·mL^-1 and 4 3μg.·mL^-1, respectively, and the highest can be as high as 980 μg.·mL^-1 and 1150 μg.·mL^-1 respectively. The ICP-MS analysis results showed that, of the analyzed trace elements, zinc has the highest concentration in both the whole solution and the water-soluble fraction, followed by Pb and Mn. ICP-MS analysis also demonstrated that Zn, Cd and As in Lanzhou PM10 were mostly in soluble forms, unlike Fe, Pb and V, which were mostly in insoluble states. The correlations between the TD20 values and the concentrations of total analyzed V, Cr, Mn, Co, Ni, Cu, Zn, As, Cd, Hg, Cs and Pb in PM10 implied that the bioreactivity of PM10 may be attributed to the water-soluble trace elements in PM10. For the whole and water soluble fractions of PM10, the individual water-soluble Zn, Mn, Pb, Fe, V, Cu, Cr, Cd, Cs and Co in Lanzhou PM10 showed an obvious negative correlation with the TD20 values, suggesting that they were probably the major trace elements responsible for the plasmid DNA damage.
