摘 要:为探讨骨形态发生蛋白2(BMP2)在体外抑制人骨肉瘤细胞株MG63的增殖和迁移并促使其凋亡的作用机制,分别用绿色荧光蛋白重组腺病毒(AdGFP)、骨形态发生蛋白2重组腺病毒(AdBMP2)感染人骨肉瘤细胞MG63,用RT-PCR和免疫细胞化学法检测核录因子κB(NF-κB)、有丝分裂素激活蛋白激酶激酶4(MKK-4)的mRNA和蛋白水平变化。结果发现,AdBMP2降低MG63中NF-κB的mRNA水平和蛋白表达,分别为对照组的38.2%和42.5%(P<0.05);但未能检测到对MKK-4表达的影响。研究表明,BMP2可以显著降低细胞中NF-κB的mRNA水平和蛋白水平,这可能是其抑制细胞增殖和迁移、促进凋亡的机制之一。To investigate the mechanism of the inhibition of BMP2 on the proliferation and migration of human osteosarcoma cell line MG63 and the promotion of MG63 apoptosis in vitro, MG63 cells were infected with AdGFP and AdBMP2, respectively and then the expressions of NF-κB and MKK-4 in mRNA level and in protein level were detected by RT-PCR and immunocytochemistry (ICC). Results show that AdBMP2 reduces the NF-κB mRNA and protein levels in MG63, but the effect on the expression of MKK-4 was not evident, which suggests that BMP2 can significantly reduce NF-κB in the mRNA level and protein level and this function may serve as the mechanism of the inhibition of BMP2 on the proliferation and migration of MG63, and the promotion of MG63 apoptosis.
