摘  要:目的提取分离山楂中表儿茶素并测定其含量。方法采用聚酰胺柱层析对山楂进行分离纯化,用反相高效液相色谱(RP-HPLC)法测定表儿茶素含量,色谱柱为DiamonsilTMSB-C18柱(200mm×4.6mm,5μm),流动相为0.02moL/LKH2PO4(pH=3.0)-乙腈(83∶17),检测波长273nm,流速1.0mL/min,柱温25℃,进样量10μL。结果表儿茶素质量浓度在0.95~15.2μg/mL范围内与峰面积线性关系良好,不同产地的山楂药材的表儿茶素含量差异较大。结论建立的方法能较好地分离并准确测定山楂中的表儿茶素。Objective To establish a determination method for the extraction and separation of epicatechin in crataegolic. Methods Epicatechin was extracted and separated from crataegolie powder which was dissolved in 30% ethanol by polyamide. A DiamonsilTM SB- C18 column (200 mm ×4.6mm, 5 μm) was used. The mobile phases was 0.02 moL/L KH2PO4 (pH =3.0)-acetonitrile(83 : 17) with the detective wavelength at 273 nm, the flow rate was 1.0 mL/min. The column temperature was 25℃ , the sample size was 10 μL. Results The content of epicatechin was from 0.95 - 15.2 μg/mL The epicatechin of hawthorn berry from different producing areas existed a great difference. Conclusion This method is simple, which can be used to determine epieatechin in crataegolic with satisfactory accuracy and reproducibility.
