摘 要:目的高脂乳剂灌胃并配合自由饮用糖水建立脂肪肝胰岛素抵抗大鼠模型。方法SD大鼠灌胃高脂乳剂及自由饮用浓度为180g.L-1的蔗糖水,每日1次,连续10wk。观察大鼠体重等一般状况;检测肝脏TG和FFA的含量;血清ALT、AST、FFA、HDL-C、LDL-C、葡萄糖(FBG)和胰岛素(FINS)水平以及肝脏的病理形态学变化。结果模型组大鼠肝脏TG、FFA含量升高,血清LDL-C、FFA、FBG、FINS含量升高,血清HDL-C降低,IRI水平升高,ISI水平降低,出现胰岛素抵抗。造模6wk末肝脏病理形态学显示出单纯性脂肪肝,8wk末为轻中度脂肪性肝炎,10wk末为中重度脂肪性肝炎。结论灌胃高脂乳剂及饮用高糖饮料10wk,可成功建立脂肪肝胰岛素抵抗大鼠模型。Aim To establish a rat model with insulin-resistant fatty liver by gastric perfusing high-fat emulsion and freely feeding sucrose solution. Methods The model SD rats were received gavage with high-fat emulsion once a day and freely fed with sucrose solution( 180 g.L^-1). The general conditions and body weight changes were observed, 10 weeks later, the hepatic TG, FFA and the serum ALT,AST, FFA, HDL-C, LDL-C, fasting glucose(FBG) , fasting insulin (FINS) were measured. And the pathological changes of liver tissues were observed. Results In the model group rats,the hepatic TG and FFA concentration increased, the serum ALT, HDL-C, FBG and FINS levels increased, whereas the serum LDL-C decreased, IRI increased and ISI decreased, so insulin resistance (IR) was created. In addition, the histopathology of liver tissues displayed that the livers presented simple fatty liver at the end of 6 weeks, mild to moderate fatty hepatitis at the end of 8 weeks and moderate to severe fatty hepatitis at the end of 10 weeks. Conclusion A rat model with insulin-resistant fatty liver can be successfully established by gastric perfusing a high-fat emulsion and freely feeding sucrose solution in 10 weeks.
