摘 要:本文在孵育的离体大鼠脊髓组织切片上,观察牛磺酸对缺氧/复氧损伤的影响。缺氧/复氧损伤引起大鼠脊髓组织ATP含量减少和LDH漏出增多,NO生成和NOS活性明显增加。L精氨酸转运Vmax增加97%(P<001),Km值无明显变化。用10和20mmol/L牛磺酸孵育与单纯缺氧/复氧组比较,显著减轻了缺氧/复氧引起的组织ATP减少和LDH漏出,NO生成分别低34%(P<001)和40%(P<001),组织L精氨酸转运Vmax降低32%(P<001)和38%(P<001),但仅轻度抑制NOS活性增加。提示牛磺酸对L精氨酸/一氧化氮系统的影响可能是其对缺氧/复氧损伤的神经保护作用的机理之一。The effect of taurine on hypoxia/reoxygenation(H/R) injury in tissue slices of isolated rat spinal cord was investigated.There are decrease of ATP content,increase of LDH leakage,No production and NO Synthase(NOS)activity were obviously elevated during H/R.Maximum transport velocity(Vmax)of L Arginine(L Arg)is 97% higher(P<0 01),while Michaelis Constant(Km)showed no significant changes.In comparison with H/R group,10 and 20 mmol taurine could significantly alleviate ATP decrease and LDH leakage;NO production reduced 34% (P<0 01) and 40%(P<0 01)respectively.Vmax of L Arg lowered 32%(P<0 01) and 38%(P<0 01).But NOS activity was slightly inhibited.Thus it might be suggested that effect of taurine on L Arg /NO sytem may be one of the nervous protective mechanisms during H/R injury.
