摘 要:目的利用流式细胞仪技术分析比较基因敲入(KIMAP)和转基因(TGMAP)小鼠前列腺癌模型。方法对来源于KIMAP和TGMAP的肿瘤标本进行了流式细胞仪分析,并与临床前列腺癌进行了比较。结果来源于TGMAP(n=8例)和KIMAP(n=9例)的小鼠前列腺癌标本流式细胞仪分析显示,多数KIMAP肿瘤为双倍体,与临床相近,而所有TGMAP肿瘤均为非整倍体。结论由于KIMAP模型成功地模仿了人类前列腺癌特征,在前列腺癌临床前期研究中具有潜在的应用价值。[Objective] This study used flow cytometry DNA analysis to measure and compare the new knock-in mouse adenocarcinoma prostate model (KIMAP) with transgenic mouse adenocareinoma prostate model (TGMAP) and human prostate cancer (CAP). [Methods] Flow cytometry DNA analysis was performed on malignant prostate tissues obtained from both TGMAP and KIMAP models and compared with human CaP. [Results]Mice with CaP from TGMAP (n =8) and KIMAP (n = 9) models showed different results. Flow cytometry analysis demonstrated that the majority of tumor tissues in KIMAP were diploid, analogous to the human condition, while all the TGMAP mice showed aneuploid tumors. [ Conclusion] Due to the effective mimicry of human CaP tumors demonstrated by the PSP-KIMAP model, this study advocates its use in the clinical study of human CaP.
