摘 要:[目的]研究游离酶和壳聚糖-海藻酸钠微胶囊固定化酶的酶学性质。[方法]以壳聚糖和海藻酸钠为载体,利用微胶囊技术固定木聚糖酶。[结果]结果表明,壳聚糖-海藻酸钠微胶囊法固定化木聚糖酶的固定率为39.4%,固定化酶和游离酶的Km值分别为0.765和0.687 g/L,固定化酶的热稳定性、重复操作稳定性和贮藏稳定性有了明显提高。[结论]采用壳聚糖-海藻酸钠微胶囊法固定化木聚糖酶具有一定的应用价值。[ Objective ] The enzymatic property of free Xylanaso and immobilized xylanase was researched. [ Method ] The sodium alginate and ehitosan being taken as carrier, the microcapsu]e technique was applied to immobilize xylanase. [ Results ] The results indicated that the recovery percentage of enzyme activity was 39.4% when the xylanase was immobilized with the sodium alginate-chitosan microcapsule, and the Km value of immobilized xylanaso and free xylanase were 0.765 and 0.687 g/L, respectively. And also, temperature stability, operation stability and storage stability of immobilized xylanase were obviously improved. [ Conclusion ] It was concluded that the method of the sodium alginate-chitosan microcapsule immobilizing xylanase had the value in industry.
