摘 要:目的:探讨前列腺癌微环境中树突状细胞(DC)分布、功能状态的变化及其与记忆性T细胞的关系。方法:选取40例前列腺增生(BPH),40例前列腺上皮内瘤(PIN)及46例前列腺癌(Pca)石蜡组织标本,用S100标记组织中总DC、CD83标记成熟DC、CD45RO标记组织中记忆性T细胞,用SP法行免疫组化染色;观察阳性细胞分布,计算阳性细胞浸润密度,分析DC与T细胞的相关性。结果:由BPH、PIN到Pca的S100+DC、CD45RO+T浸润密度逐渐降低,前列腺癌组织降低明显(P<0.01);Pca中CD83+DC浸润密度高于BPH和PIN(P<0.05)。前列腺癌组织中,S100+DC主要分布在肿瘤间质(P<0.01),CD83+DC主要分布在癌上皮间及癌周(P<0.01),且CD83+DC与CD45RO+T浸润密度呈正相关(rs=0.831,P<0.05)。结论:前列腺癌组织中,DC数量减少且存在功能缺陷,不同功能状态的DC存在区域性分布;成熟DC数量的减少可能导致T细胞免疫功能下降。Objective:To investigate the distribution and functional status of dendritic cells (DC)in the microenvironment of prostate cancer, and its relationship with memory T lymphocyte. Methods:Forty benign prostatic hyperplasia (BPH),40 prostatic intraepithelial neoplasm (PIN)and 46 prostate cancer (Pca) specimens were selected for immunohistochemistry (SP method). The infiltrating density of the S100+DC, CD83 +DC and CD45RO +T lymphocyte were calculated, meanwhile their distribution were observed. The relationship between DC and T lymphocyte was statistically compared. Results: The infiltrating density of S100+DC and CD45RO+T lymphocyte decreased gradually from BPH, PIN to Pca, especially in prostate cancer (P 〈 0.01). While, in prostate cancer, the infiltrating density of CD83+DC was more than that of BPH and PIN (P 〈 0.05). S100+DC mainly localized in the tumor stroma (P 〈 0.01). CD83+DC mainly presented in the tumor epithelium and advancing tumor margin (P 〈 0.01 )and directly contacted with tumor cells. There was a significant correlation between the infiltrating density of CD83+DC and T lymphocytes in Pca (rs=0.831 ,P 〈 0.05). Conclusion: The number of DC decreased in prostate cancer. The different DC had different distributions in Pca and the lack of activated DC may decrease the function of T lymphocyte.
