摘 要:目的:研究杜仲叶醇提取物(alcohol extractive of Folium Eucommiae,AEFE)对冠状动脉粥样硬化心脏病的保护作用及机制。方法:高脂饮食加维生素D3(总量60万u/kg,分3天腹腔注射)建立大鼠冠状动脉粥样硬化(Atherosclerosis,AS)模型。AEFE以70mg/kg、140mg/kg及420mg/kg每天灌胃给药6周。生化方法测定血浆脂质(TC、TG、HDL、LDL和apoB/apoAI)、MDA和NO含量以及SOD活性;免疫组织化学法检测ICAM-1和TNF-α在冠脉和心脏的表达情况;光镜下观察冠状动脉及心肌的病理改变。结果:AEFE140mg/kg和420mg/kg剂量明显减轻冠状动脉粥样硬化病变程度和心肌的损伤,下调冠脉和心脏ICAM-1蛋白的表达;同时,AEFE各剂量可明显降低TC、TG、LDL、apoB/apo(A-I)和MDA浓度,升高HDL、NO浓度和SOD活性;但不影响TNF-α蛋白在冠脉和心肌的表达。结论:AEFE可以通过降血脂、抗氧化损伤、促进NO释放和抑制ICAM-1蛋白的表达,阻遏冠状动脉粥样硬化心脏病的发生和发展。Objective: To investigate the anti-atherosclerosis effects of alcohol extractive of Folium Eucommiae (AEFE) and its mechanism of protective effect on coronary atherosclerotic heart disease through experimental atheroselerosis. Methods: In vivo, experimental atherosclerosis in rats was produced by feeding with atherogenic-diet and vitaminD3 administration ( 200,000 IU/kg by intraperitoneal injected on 3 successive days) for 8 weeks. AEFE (70,140 and 420mg/kg. d ,ig) was given for 6 weeks. Then, the effects of different AEFE dosage and lovastatin on the following parameters were evaluated by biochemistry method and immunohistochemistry, which were levels of plasma lipid profiles, SOD, MDA, NO, as well as expression of ICAM-1 and TNF-α on coronary artery and heart. Results: AEFE 140 and 420 mg/kg obviously ameliorated the pathologic changes and inhibited the expression of ICAM-1 on coronary artery and myocardium, but did not influence the expression of TNF-α . At the same time, all of dosages of AEFE not only significantly decrease the elevations of levels in TC, TG and LDL, ApoB/ApoAI ratio, but also reverse the decrease in HDL and NO. The increase of MDA levels and decrease of SOD activity which induced by atherogenic-diet were reversed after treatment with AEFE. Conclusion: The results suggest that the AEFE show a promising protective effect on coronary atherosclerotic heart disease and its protection may involve in regulating plasma lipid metabolic disorder, enhancing anti-ox- idant capacity, releasing NO from endothelium and inhibiting the expression of ICAM-1 .
