胰腺癌神经浸润转移体外模型构建的初步探讨

摘  要:目的使用旋转细胞培养系统(rotating cell culture system,RCCS)模拟微重力环境,建立胰腺癌神经转移的体外模型。方法将裸鼠背根神经节(dorsal root ganglion,DRG)和人胰腺癌细胞株MIA PaCa-2置于RCCS系统中混合培养,在额定时间点取神经组织送病理,Gomori染色观察神经组织结构和胰腺癌细胞在神经组织块中生长情况。结果DRG培养12h后,镜下可见有一神经突起自DRG周缘长出;72h后,可见神经突起显著延长、增多,多数突起结构有一分支,大体呈放射状排列。MIA PaCa-2细胞能够溶解基质。癌细胞与DRG共培养48h,可见有癌细胞靠近神经纤维并贴附其生长。72h可见癌细胞形成包绕神经纤维的细胞团,典型的表现为形成以神经纤维为中轴的纺锤样结构。结论使用RCCS系统模拟微重力环境,构建胰腺癌神经转移体外模型,可以用于胰腺癌神经转移过程和机制的研究,为研究胰腺癌细胞和神经组织间的相互关系提供新的方法。Objective To establish a novel experimental nerve metastasis model of human pancreatic cancer in vitro using the rotating cell culture system (RCCS) to simulate micro-gravity culture. Methods Nude mouse normal nerve tissues and human pancreatic cancer cell line MIA PaCa-2 were co-cultured in the micro-gravity environment simulated by RCCS. The human pancreatic cancer MIA PaCa-2 cells and nude mouse nerve tissue explants were maintained in DMEM/F-12 supplemented with 10% fetal calf serum and incubated at 37℃ in a 5% CO2 incubator. The vessel was rotated at a speed of 16 rpm so that the nerve tissue could be maintained in the state of suspension. The culture medium was replaced every day.The samples were taken and medium replaced under sterile conditions. The samples were stained with Gomori for observation of growth of pancreatic cancer cells in the nerve tissue. Results One nerve tuber outgrow from peripheral DRG after cultured at 12 hours and nerve tuber remarkably prolonged and manifolded at 72 hours. MIA PaCa-2 could dissolve Matrigel. The pancreatic cancer cells adhered to and grew on the DRG surface at 48 hours, massively grew and formed metastatic colonies in the DRG at 72 hours after their incubation in the culture medium. Conclusions This model provides a new tool for investigating the mechanism of nerve metastasis of pancreatic cancer.
