摘 要:目的探讨单层细胞角蛋白CK19mRNA的表达与口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)的局部侵袭与转移的关系。方法选用23例OSCC组织及相应癌旁组织,通过半定量RT-PCR方法观察OSCC中CK19-mRNA的表达,探讨其与肿瘤临床分期、分化程度、淋巴结转移的关系。检测CK19-mRNA在OSCC患者外周血中的表达,探讨其与OSCC远处转移及复发的关系。结果23例OSCC组织中CK19-mRNA均呈阳性表达,且表达量明显高于癌周组织(P<0.01)。CK19-mRNA表达仅与分化程度有关。23例OSCC手术前后外周血中均无CK19-mRNA的表达。结论CK19-mRNA可作为辅助诊断OSCC分化程度的指标,其表达水平的改变与OSCC转移无关,不能作为判断OSCC远处转移的指标。Objective To investigate the relationship between expression of CK19 and invasion and metastasis of oral squamous cell carcinoma. Methods Twenty-three patients with oral squamous cell carcinoma were investigated for the expression of CK19-mRNA by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). Correlations between clinical stages, pathological differentiation, lymphatic metastasis and the expression of CK19-mRNA were evaluated. CK19-mRNA expression in peripheral blood of these patients and 23 healthy people were also examined. During follow-up after operation,the peripheral blood was collected again for the expression of CK19-mRNA. Results Expression of CK19-mRNA was found in all patients, but the expression level was different. The expression of CK19-mRNA was only associated with tumor differentiation. CK19-mRNA was negative in all of 23 blood specimens before operation and during follow-up. Conclusion CK19-mRNA is correlated with tumor differentiation of OSCC and not associated with tumor metastasis.
