摘  要:目的探讨血浆同型半胱氨酸(Hcy)水平与ApoE基因多态性在阿尔茨海默病(AD)发病机制中的关系。方法采用循环酶法及聚合酶链式反应-限制性片段长度多态性法(PCR-RFLP)检测145例AD患者血浆同型半胱氨酸水平与ApoE基因多态性分布。结果患者血浆Hcy水平与正常对照比较,差异显著(P<0.01);含2个ε4等位基因的患者血浆Hcy水平较不含ε4等位基因及含1个ε4等位基因的显著增高,差异有统计学意义(P<0.01)。结论AD患者血浆Hcy水平与ApoEε4基因剂量密切相关,遗传是导致患者血浆Hcy水平增高的一个重要原因。Objective To explore the relationship between plasma homocysteine (Hey) levels and apolipoprotein E (APOE) gene polymorphism in Alzheimer's disease (AD). Methods The plasma Hcy levels and the distribution of ApoE gene polymorphisms were observed with the method of enzymatic cycling assay and polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP) in 145 patients with AD (AD group). Results There were significant difference in plasma Hey levels between the AD group and the control group (P 〈 0. 01 ). There were significant difference in plasma Hey levels between two ε4 allele contained, none ε4 allele contained and one allele contained in AD group (P 〈 0. 01 ). Conclusions The plasma homocysteine levels is closely associated with the ε4 gene dosage of apolipoprotein E, heredity is one of the most important reasons which result in high plasma levels.
