摘 要:目的观察COM.TEC型血细胞分离机MNC程序采集外周血造血干细胞效果。方法用COM.TEC型血细胞分离机MNC程序采集16例血液系统肿瘤患者外周血干细胞,患者采用化疗、粒细胞集落刺激因子动员,采集的干细胞用流式细胞仪检测CD34+细胞数。共采集32次,每例采集2次,每次处理抗凝全血(9158±2166)mL,抗凝剂(919.78±230.60)mL,时间(178.1±38.0)min。结果采集1次CD34+细胞数为(1.62±0.32)×106/kg,MNC采集效率为(81.25±14.57)%,单采后患者血小板(plt)、血红蛋白(Hb)、红细胞比容(Hct)分别下降13.0%、7.2%、9.1%,与采集前比较,差异无统计学意义(P>0.05)。采集过程中未发生严重不良反应。结论COM.TEC型血细胞分离机MNC程序能采集到血小板含量低、产量高而有效的自体CD34+细胞,采集方法可靠,可以满足临床治疗要求。Objective To observe the efficiency of Fresenius COM. TEC MNC process in collection of peripheral hematopoietic stem cell. Methods The Fresenius COM. TEC blood cell separator was utilized to collect hematopoietie stem cell from 16 patients which suffer from neoplastic hematologic disorder. The patients were treated with chemo add G-CSF arouse. The number of CD34^+ cell antigent presentation were detected by flow cytometry. With 16 patient 32 times collect and each patient 2 times. Disposal anticoagulation blood(9 158±2 166)mL. Time(178. 1±38)min. Decoagulant(919.78±230.6)mL. Results The number of CD34^+cell was(1.62±0.32) )〈 106/kg and MNC collection efficiency was(81.25 ± 14.57)% after one time gathered. Ph, Hb, Hct had reduced 13% ,7.2% ,9.12%. There were no severe adverse reaction at all the process. Conclusion COM. TEC blood cell separator automatic MNC process can successfully collect CD34 cell high output efficienily with low platelet. The collection method is reliable and achieve clinical request.
