摘 要:目的初步探讨代谢性核受体在人结肠癌组织中的表达及意义。方法分别用RT-PCR和Western blot检测维甲酸受体α(RARα)、孕烷X受体(PXR)、维生素D受体(VDR)、过氧化物酶体增殖体激活受体α(PPARα)和法尼酯衍生物X受体(FXR)在17例结肠癌组织中的表达,并进行半定量统计分析。结果与相应正常组织比较,除FXR外,结肠癌组织中RARα、PXR、VDR和PPARαmRNA水平均显著上调,差异有统计学意义(P<0.05);而结肠癌组织RARα和PXR蛋白表达水平也显著高于相应正常组织,差异有统计学意义(P<0.05)。结论结肠癌中RARα、PXR、VDR和PPARα表达上调可能与结肠癌发生发展相关。Objective To investigate the expression of metabolic nuclear receptors in colon cancer. Methods The mRNA and nuclear protein levels of RARa,PXR,VDR,PPARa and FXR in colon cancer and matched normal tissues were determinded by RTPCR and western blot. Band intensity was quantified using Quantity One 4.2.0 software and datas were analysised by Paired-Samples T Test. Results The mRNA expression of RARα,PXR,VDR and PPARα was significantly upregulated (P〈0.05) in 17 colon cancer tissues compared with the matched normal colon tissues except for FXR. Furtherly, the nuclear protein expression of RARα and PXR also wassignificantly increased in colon cancer tissues (P〈0.05). Conclusion The differential upregulation of RARα,PXR,VDR and PPARα may be associated with the colon cancer cancerogenesis and development.
